【摘要】：目的探討沉默信息調(diào)節(jié)因子1(SIRT1)基因rs12778366位點(diǎn)和載脂蛋白C3(APOC3)基因rs2854116位點(diǎn)單核苷酸多態(tài)性(SNP)與非酒精性脂肪性肝病(NAFLD)發(fā)病易感性的關(guān)系。方法數(shù)據(jù)源于2015年蘇州市相城區(qū)醫(yī)院體檢人群,共收集病例560人,根據(jù)B超結(jié)果,篩選納入NAFLD病例132例,隨機(jī)選取正常對(duì)照人群252例,采集血液樣本,分析總膽固醇、甘油三酯、高密度脂蛋白膽固醇、低密度脂蛋白膽固醇、空腹血糖、丙氨酸氨基轉(zhuǎn)移酶、天門冬氨酸氨基轉(zhuǎn)移酶和血尿酸的血生化指標(biāo),及身高、體重、腰圍、臀圍、血壓一般指標(biāo),全血中提取DNA,采用聚合酶鏈反應(yīng)(PCR)、Mass ARRAY時(shí)間飛行質(zhì)譜技術(shù)分析SIRT1 rs12778366和APOC3rs2854116位點(diǎn)基因型。結(jié)果 SIRT1 rs12778366位點(diǎn)TC+CC基因型攜帶者相對(duì)于TT基因型攜帶者,發(fā)生NAFLD的OR=1.126(95%CI 0.673~1.886),P0.05。與APOC3 rs2854116位點(diǎn)CC基因型攜帶者相比較,TC+TT基因型攜帶者發(fā)生NAFLD的OR=1.044(95%CI 0.601~1.814),P0.05。校正性別、年齡、體質(zhì)指數(shù)的混雜因素后OR均無明顯改變(P0.05)。多因素Logistic回歸分析顯示:甘油三酯、體質(zhì)指數(shù)、空腹血糖、腰圍和尿酸是NAFLD發(fā)病可能的獨(dú)立危險(xiǎn)因素(P0.05),而SIRT1 rs12778366和APOC3 rs2854116位點(diǎn)基因多態(tài)性與NAFLD的發(fā)病風(fēng)險(xiǎn)無明顯關(guān)聯(lián)。結(jié)論 SIRT1 rs12778366和APOC3 rs2854116位點(diǎn)基因多態(tài)性與NAFLD的發(fā)生無明顯相關(guān)性,均不會(huì)影響NAFLD的發(fā)病風(fēng)險(xiǎn)。
[Abstract]:Objective to investigate the relationship between the single nucleotide polymorphisms (SNPs) of rs12778366 locus and apolipoprotein C3 (APOC3) gene rs2854116 locus of silencing signaling factor 1 sirt1) gene and the susceptibility to NAF LDD of non-alcoholic fatty liver disease.Methods data were collected from a total of 560cases collected from the physical examination population of Xiangcheng District Hospital of Suzhou City in 2015. According to the results of B-mode ultrasound, 132 cases of NAFLD were selected and 252 cases of normal controls were randomly selected to collect blood samples and analyze total cholesterol (TC).Serum biochemical indexes of triglycerides, high density lipoprotein cholesterol, low density lipoprotein cholesterol, fasting blood glucose, alanine aminotransferase, aspartate aminotransferase and serum uric acid, and height, weight, waist circumference, hip circumference,The genotypes of SIRT1 rs12778366 and APOC3rs2854116 loci were analyzed by polymerase chain reaction (PCR) mass ARRAY time-flight mass spectrometry (TF-MS).Results TCCC genotype carriers at SIRT1 rs12778366 locus were significantly higher than TT genotype carriers (P 0.05).Compared with CC genotype carriers of APOC3 rs2854116 locus, the OR=1.044(95%CI 0.601 and 1.814P0.05 of TCTT genotype carriers developed NAFLD.After adjusting for the mixed factors of sex, age and BMI, the OR had no significant change (P 0.05).Multivariate Logistic regression analysis showed that triglyceride, body mass index, fasting blood glucose, waist circumference and uric acid were independent risk factors for NAFLD, while SIRT1 rs12778366 and APOC3 rs2854116 locus polymorphism were not significantly associated with the risk of NAFLD.Conclusion there is no significant correlation between the polymorphism of SIRT1 rs12778366 and APOC3 rs2854116 loci and the occurrence of NAFLD, and neither of them will affect the risk of NAFLD.
