免疫共沉淀聯(lián)合質(zhì)譜分析篩選克羅恩病中Intelectin-1相互作用蛋白的初步研究

【摘要】:目的對比克羅恩病患者的病變與正常腸黏膜,篩選與腸凝集素-1(Intelectin-1)相互作用的差異蛋白,探討Intelectin-1及其相互作用蛋白在克羅恩病發(fā)展中的作用。方法利用免疫共沉淀技術(shù)篩選克羅恩病患者病變及正常組織中與Intelectin-1相互作用的蛋白;利用MALDI-TOF/TOFMS質(zhì)譜分析技術(shù)鑒定與Intelectin-1相互作用蛋白;并使用免疫共沉淀和Western blot分析技術(shù)驗證所鑒定的Intelectin-1相互作用蛋白。結(jié)果利用免疫共沉淀聯(lián)合質(zhì)譜分析技術(shù)成功篩選出4個與Intelectin-1相互作用差異蛋白,包括:腫瘤壞死因子受體相關(guān)因子3(TRAF3)、腺苷酸三磷酸酶(ATPase)、熱休克蛋白90(HSP90)、鋅指蛋白(ZNF)。結(jié)論Intelectin-1可能通過與TRAF3、ATPase、HSP90、ZNF蛋白相互作用,而影響克羅恩病的發(fā)展。
[Abstract]:Objective to compare the pathological changes with normal intestinal mucosa in patients with Crohn's disease and to screen the differential proteins interacting with lectin-1 and Intelectin-1. To investigate the role of Intelectin-1 and its interacting proteins in the development of Crohn's disease. Methods Immunoprecipitation technique was used to screen the proteins interacting with Intelectin-1 in pathological changes and normal tissues of patients with Crohn's disease. The protein interacting with Intelectin-1 was identified by MALDI-TOF/TOFMS mass spectrometry. The identified Intelectin-1 interacting proteins were verified by immunoprecipitation and Western blot analysis. Results four differentially interacting proteins with Intelectin-1 were successfully screened by immunoprecipitation and mass spectrometry. These include: tumor necrosis factor receptor-associated factor 3TRAF3, adenylate triphosphatase (ATP), heat shock protein 90 (HSP90), zinc finger protein (ZNFN). Conclusion Intelectin-1 may affect the development of Crohn's disease by interacting with the protein of TRAF3, ATPase and HSP90.
