【摘要】：目的： 1、了解喉鱗狀細(xì)胞癌（簡稱喉鱗癌）患者外周血清中抗survivin抗體和survivin mRNA的相關(guān)性。 2、比較抗survivin抗體、抗p53抗體和抗c-myc抗體作為生物標(biāo)志物在喉鱗癌檢出中的作用，分析聯(lián)合檢測的價(jià)值。 3、探討血清生物標(biāo)志物在喉鱗癌早期診斷及發(fā)生、發(fā)展和預(yù)后監(jiān)測方面的臨床意義。 方法： 1、酶聯(lián)免疫吸附試驗(yàn)(ELISA)用于檢測血清樣本中抗survivin抗體、抗p53抗體和抗c-myc抗體的表達(dá)。 2、半定量聚合酶鏈?zhǔn)椒磻?yīng)（RT-PCR）檢測相同樣本中survivinmRNA的表達(dá)。 結(jié)果： 1、抗survivin抗體和survivin mRNA在喉鱗癌患者外周血清中高表達(dá)，，特別是有頸淋巴結(jié)轉(zhuǎn)移的樣本，且二者的表達(dá)水平顯著相關(guān)。 2、抗p53抗體和抗c-myc抗體在相同樣本中同樣高表達(dá)。 3、在接受根治性手術(shù)治療后抗survivin抗體與survivin mRNA的表達(dá)水平顯著下降，抗p53抗體和抗c-myc抗體得到了類似的結(jié)果。 4、抗survivin抗體、抗p53抗體和抗c-myc抗體單獨(dú)檢測時(shí)均獲得了較高的陽性率，三種抗體聯(lián)合檢測的陽性率并不優(yōu)于抗survivin抗體的陽性率，但優(yōu)于抗p53抗體和抗c-myc抗體。 結(jié)論： 1、喉鱗癌外周血清中抗survivin抗體與survivin mRNA的表達(dá)水平相關(guān)。 2、抗survivin抗體可能是喉鱗癌的一種有價(jià)值的生物標(biāo)志物。Survivin在喉鱗癌的發(fā)生、發(fā)展進(jìn)程中可能扮演重要角色。 3、聯(lián)合檢測三種不同的腫瘤標(biāo)志物并不能有效提高喉鱗癌的術(shù)前檢出率，但可能在監(jiān)測手術(shù)效果方面具有積極意義。

[Abstract]:Objective: 1. To investigate the correlation between anti-survivin antibody and survivin mRNA in peripheral serum of patients with laryngeal squamous cell carcinoma (LSC). 2. To compare the role of anti-survivin antibody, anti-p53 antibody and anti-c-myc antibody as biomarkers in the detection of laryngeal squamous cell carcinoma, and to analyze the value of combined detection. 3. To investigate the clinical significance of serum biomarkers in early diagnosis, occurrence, development and prognosis monitoring of laryngeal squamous cell carcinoma. Methods: 1. Enzyme-linked immunosorbent assay (ELISA) was used to detect the expression of anti-survivin antibody, anti-p53 antibody and anti-c-myc antibody in serum samples. 2. Semi-quantitative polymerase chain reaction (RT-PCR) was used to detect the expression of survivinmRNA in the same sample. Results: 1. Anti-survivin antibody and survivin mRNA were highly expressed in peripheral serum of patients with laryngeal squamous cell carcinoma, especially in samples with cervical lymph node metastasis, and their expression levels were significantly correlated. 2. Anti-p53 antibody and anti-c-myc antibody were also highly expressed in the same sample. 3. The expression of anti-survivin antibody and survivin mRNA decreased significantly after radical operation, and similar results were obtained between anti-p53 antibody and anti-c-myc antibody. 4. The positive rates of anti-survivin antibody, anti-p53 antibody and anti-c-myc antibody were high when detected alone. The positive rate of the three antibodies was not better than that of anti-survivin antibody, but better than that of anti-p53 antibody and anti-c-myc antibody. Conclusion: 1. The expression of anti-survivin antibody in peripheral serum of laryngeal squamous cell carcinoma is correlated with the expression of survivin mRNA. 2. Anti-survivin antibody may be a valuable biomarker of laryngeal squamous cell carcinoma. Survivin may play an important role in the occurrence and development of laryngeal squamous cell carcinoma. 3. The combined detection of three different tumor markers can not effectively improve the preoperative detection rate of laryngeal squamous cell carcinoma, but it may have positive significance in monitoring the surgical effect.
