【摘要】：目的 研究大鼠玻璃體腔內(nèi)注射尿激酶(UK)對視網(wǎng)膜血管通透性的影響。 方法 SD健康大鼠30只,隨機(jī)分為2組,每組15只(15眼)。實(shí)驗(yàn)組大鼠右眼玻璃體腔內(nèi)注射UK350U/4u l,對照組大鼠右眼玻璃體腔內(nèi)注射0.01M磷酸鹽緩沖液(PBS)4μ l。24小時(shí)后大鼠尾靜脈注射追蹤劑依文思藍(lán)(EB),待EB循環(huán)2小時(shí)后,摘除大鼠右眼球,取出玻璃體,使用間接酶聯(lián)免疫吸附試驗(yàn)(ELISA)檢測玻璃體內(nèi)EB吸光度。根據(jù)EB濃度-吸光度的標(biāo)準(zhǔn)曲線,獲得大鼠玻璃體腔內(nèi)EB的質(zhì)量濃度,采用SPSS17.0對數(shù)據(jù)進(jìn)行統(tǒng)計(jì)學(xué)分析。 結(jié)果 1.統(tǒng)計(jì)學(xué)軟件制作依文思藍(lán)的濃度-吸光度標(biāo)準(zhǔn)曲線可以看出：依文思藍(lán)的濃度在0-30μ g/ml,吸光度在0.008-0.12的范圍內(nèi)線性關(guān)系良好,回歸方程為y=0.0034x+0.0044,R2=0.9941,n=6。 2.顯微鏡下觀察大鼠右眼球,可見實(shí)驗(yàn)組大鼠眼球顏色明顯藍(lán)于對照組大鼠眼球。實(shí)驗(yàn)組大鼠眼球普遍為藍(lán)色,個(gè)體間顏色差異較��；對照組大鼠眼球基本上無色,少數(shù)眼球稍藍(lán)。 3. ELISA法測得大鼠玻璃體內(nèi)EB的吸光度結(jié)果顯示：實(shí)驗(yàn)組大鼠玻璃體腔內(nèi)EB平均吸光度值為(0.0558±0.0112)/25μ l,對照組大鼠玻璃體腔內(nèi)EB的平均吸光度值為(0.0145±0.0077)/25μ l,兩組差異有統(tǒng)計(jì)學(xué)意義(t=11.746,P=0.000)。 4.由EB濃度-吸光度標(biāo)準(zhǔn)曲線得出大鼠玻璃體內(nèi)EB的平均質(zhì)量濃度結(jié)果顯示：實(shí)驗(yàn)組大鼠玻璃體內(nèi)EB的平均質(zhì)量濃度為(15.1078±3.2905)u g/25μ l,對照組大鼠玻璃體腔內(nèi)EB的平均質(zhì)量濃度為(3.3093±2.1615)μ g/25u l,二組差異有統(tǒng)計(jì)學(xué)意義(t=11.607,P=0.000)。 結(jié)論 1、大鼠玻璃體腔內(nèi)注射UK可以增大視網(wǎng)膜血管的通透性 2、EB可以作為一種測量視網(wǎng)膜血管通透性的理想追蹤劑
[Abstract]:Objective to study the effect of intravitreal injection of urokinase (UK) on retinal vascular permeability in rats. Methods 30 healthy SD rats were randomly divided into two groups with 15 eyes in each group. Rats in the experimental group received intravitreal injection of UK350U/4u-1 in the right eye, while in the control group, 0.01m phosphate buffer solution (PBS) was injected into the vitreous cavity of the control group for 24 hours after injection of the tracking agent Evans blue (EB), into the tail vein of the rats. After 2 hours of EB circulation, the right eyeball of the rat was removed, the vitreous body was removed, and the EB absorbance in the vitreous was detected by indirect enzyme-linked immunosorbent assay (ELISA). According to the standard curve of EB concentration-absorbance, the mass concentration of EB in rat vitreous cavity was obtained, and the data were analyzed statistically by SPSS17.0. Outcome 1. The standard curve of concentration-absorbance of Evans blue produced by statistical software shows that the linear relationship between the concentration of Evans blue is 0-30 渭 g / ml, the absorbance is 0. 008 渭 g / ml, and the regression equation is y = 0. 0034 4 x 0. 0044, and the regression equation is y = 0. 0034 x 0. 0044. R _ (2), n ~ (2) 0. 941, n (?) 6. 2. The right eyeball of the rats was observed under microscope. The eye color of the experimental group was obviously blue than that of the control group. The eyes of the experimental group were generally blue with little color difference among individuals, while those of the control group were basically colorless and a few of the eyeballs were a little blue. 3. The absorbance of rat vitreous EB measured by ELISA showed that the average absorbance value of EB in vitreous cavity of experimental group was (0.0558 鹵0.0112) / 25 渭 l, and the absorbance of EB in vitreous cavity was (0.0558 鹵0.0112) / 25 渭 l in experimental group. The average absorbance of EB in vitreous cavity of the control group was (0.0145 鹵0.0077) / 25 渭 l, and there was a significant difference between the two groups (t = 11.746, P = 0.000). 4. According to the EB concentration-absorbance standard curve, the average mass concentration of EB in the vitreous of rats in the experimental group was (15.1078 鹵3.2905) ug / 25 渭 l, the average concentration of EB in the vitreous of the experimental group was (15.1078 鹵3.2905) ug / 25 渭 l. The mean mass concentration of EB in vitreous cavity of the control group was (3.3093 鹵2.1615) 渭 g / L, and there was a significant difference between the two groups (t = 11.607, P = 0.000). Conclusion 1. Intravitreal injection of UK can increase retinal vascular permeability 2. EB can be used as an ideal tracer to measure retinal vascular permeability.
