甲狀腺相關(guān)眼病患者外周血單個(gè)核細(xì)胞中微小RNA-146a的異常表達(dá)及意義

【摘要】:目的探討微小RNA-146a (microRNA-146a, miRNA-146a, miR-146a)在甲狀腺相關(guān)眼病(TAO)患者外周血單個(gè)核細(xì)胞中的表達(dá)情況及臨床意義,為TAO的治療提供新思路。 方法收集TAO患者、Gaves病(GD)不伴眼病患者和正常人對(duì)照者空腹靜脈全血標(biāo)本各20例,分離各組患者外周血單個(gè)核細(xì)胞(PBMCs),用TRIzol提取PBMCs的總miRNA,再以頸環(huán)結(jié)構(gòu)的引物逆轉(zhuǎn)錄出相應(yīng)的DNA,然后以miR-146a的PCR引物進(jìn)行實(shí)時(shí)熒光定量PCR(qRT-PCR)檢測(cè)各組標(biāo)本PBMCs中miR-146a的表達(dá)情況,電泳觀察結(jié)果. 結(jié)果與正常人相比,TAO患者和GD不伴眼病患者PBMCs中miR-146a相對(duì)表達(dá)量明顯降低(P0.05),下調(diào)率分別為98.02%和67.60%;較GD不伴眼病患者,miR-146a在TAO患者PBMCs中相對(duì)表達(dá)量更低(P0.05),下調(diào)率差值為30.40%。 結(jié)論1.基于SYBR Green Ⅰ染料熒光分析的莖環(huán)qRT-PCR方法檢測(cè)miR-146a表達(dá)的特異性良好; 2、miR-146a的表達(dá)下調(diào)可能參與了TAO的發(fā)病機(jī)制,為進(jìn)一步研究微小RNA在TAO發(fā)病機(jī)制中的作用打下基礎(chǔ)。

Results Compared with normal subjects, the relative expression of miR-146a in patients with TAO and GD was significantly lower than that of normal subjects (P0.05). The down-regulation rate was 98.02% and 67.60%, respectively. The relative expression of miR-146a in PBMCs of patients with GD was lower (P0.05), and the difference of down-regulation was 30. 40%. Conclusion 1. The expression of miR-146a is detected by the stem-loop qRT-PCR method based on the fluorescence analysis of SYBR Green I dye. Good specificity; the down-regulation of miR-146a may be involved in the pathogenesis of TAO, in order to further study the mechanism of small RNA in the pathogenesis of TAO
