HMGA1與Ezrin在喉癌組織中的表達(dá)及相關(guān)性研究

【摘要】:目的:探討高遷移蛋白A1(high mobility group protein A1,HMGA1)與埃茲蛋白(Ezrin)在喉癌組織中的表達(dá)及關(guān)系。 方法:采用免疫組織化學(xué)(SP法)染色方法檢測(cè)HMGA1、Ezrin蛋白在50例喉癌組織、30例喉不典型增生組織、30例喉息肉中的表達(dá);采用實(shí)時(shí)定量PCR法測(cè)定法檢測(cè)HMGA1、Ezrin mRNA在30例新鮮喉癌組織及30例癌切緣組織的表達(dá);Western Blot方法檢測(cè)HMGA1蛋白及Ezrin蛋白在30例新鮮喉癌組織及30例癌切緣組織的表達(dá),分別統(tǒng)計(jì)分析二者表達(dá)與臨床病理特征的關(guān)系及相關(guān)性。 結(jié)果:(1)HMGA1在喉息肉、喉不典型增生組織和喉癌組織的表達(dá)陽(yáng)性率分別為13.0%(4/30)、53.0%(16/30)和68.0%(34/50),三組比較有統(tǒng)計(jì)學(xué)差異(P 0.05,表2)。HMGA1蛋白表達(dá)與喉癌組織的臨床分期、組織學(xué)分級(jí)及轉(zhuǎn)移密切相關(guān)(P0.05,表3)。Ezrin蛋白在對(duì)應(yīng)三組陽(yáng)性率分別為23.3%(7/30),50.0%(15/30)和64.0%(32/50),三組比較有統(tǒng)計(jì)學(xué)差異(P 0.05)。(2)實(shí)時(shí)定量PCR示:在30例喉癌組織(100%)以及配對(duì)癌切緣組織(100%)中均檢測(cè)到HMGA1的表達(dá),兩者在喉癌組織相對(duì)于癌切緣組織的表達(dá)量具有顯著差異(HMGA1: P 0.01;Ezrin: P 0.05)。(3) Western Blot提示:在30例喉癌組織(100%)以及配對(duì)癌切緣組織(100%)中均檢測(cè)到HMGA1蛋白及Ezrin蛋白的表達(dá),兩者在喉癌組織相對(duì)于癌切緣組織的表達(dá)量具有顯著差異,(HMGA1, P 0.01;Ezrin, P0.01),單因素方差分析示,HMGA1、Ezrin與組織學(xué)分級(jí)(P 0.05)、淋巴結(jié)轉(zhuǎn)移(P<0.05)、臨床分期(P 0.05)及淋巴結(jié)轉(zhuǎn)移有關(guān)(P 0.05)有關(guān),而與吸煙、年齡及性別無(wú)關(guān)(P0.05)。(4)喉癌組織中HMGA1及Ezrin蛋白、mRNA水平表達(dá)正相關(guān)(實(shí)時(shí)定量PCR:r=0.385,P=0.000;免疫組織化學(xué):x2=7.172,r=0.379,P=0.012;Western blot:r=0.412,,P=0.000)。 結(jié)論:(1)HMGA1和Ezrin蛋白及mRNA在喉癌組織中表達(dá)異常,其表達(dá)量高于正常喉上皮組織;在喉不典型增生惡性轉(zhuǎn)化過(guò)程中,隨病變程度增加,HMGA1、Ezrin表達(dá)陽(yáng)性率逐漸增加;(2)HMGA1和Ezrin蛋白及mRNA與淋巴結(jié)轉(zhuǎn)移、病理分級(jí)和臨床分期相關(guān),與患者的年齡、性別、腫瘤的部位無(wú)明顯相關(guān),兩者在喉癌組織的表達(dá)存在正相關(guān);(3) HMGA1和Ezrin在喉癌組織中的表達(dá)增高,在喉癌的發(fā)生、發(fā)展過(guò)程中可能起協(xié)同作用,可作為喉癌預(yù)測(cè)轉(zhuǎn)移和判斷預(yù)后的生物學(xué)標(biāo)志物。
[Abstract]:Aim: to investigate the expression and relationship between high migration protein A1 (high mobility group protein A1 (HMG A1) and Ezrin (Ezrin) in laryngeal carcinoma. Methods: immunohistochemical staining (SP method) was used to detect the expression of HMGA1,Ezrin protein in 50 cases of laryngeal carcinoma, 30 cases of laryngeal dysplasia and 30 cases of laryngeal polyps. Real-time quantitative PCR was used to detect the expression of HMGA1,Ezrin mRNA in 30 fresh laryngeal carcinoma tissues and 30 incisor margin tissues. Western Blot method was used to detect the expression of HMGA1 protein and Ezrin protein in 30 fresh laryngeal carcinoma tissues and 30 incisor margin tissues. The correlation between the expression and clinicopathological features was analyzed statistically. Results: (1) the positive rates of HMGA1 expression in laryngeal polyp, laryngeal dysplasia and laryngeal carcinoma were 13.0% (4 / 30), 53.0% (16 / 30) and 68.0% (34 / 50), respectively. The expression of HMGA1 protein was correlated with the clinical stage, histological grade and metastasis of laryngeal carcinoma (P 0.05, P < 0.01). The positive rates of Ezrin protein in the corresponding three groups were 23.3% (7 / 30), 50.0% (15 / 30) and 64.0% (32 / 50), respectively. There was statistical difference among the three groups (P0.05). (2). The expression of HMGA1 was detected in 30 cases of laryngeal carcinoma (100%) and matched adjacent tissues (100%), and the expression of PCR was detected in 30 cases of laryngeal carcinoma (100%) and in matched adjacent tissues (100%). There was a significant difference in the expression of HMGA1: P 0.01 between laryngeal carcinoma and tumor incisors (P < 0.01). Ezrin: P 0.05). (3) Western Blot showed that the expression of HMGA1 protein and Ezrin protein were detected in 30 cases of laryngeal carcinoma (100%) and in matched adjacent tissues (100%), and the expression of HMGA1 protein and Ezrin protein were detected in 30 cases of laryngeal carcinoma (100%). There was a significant difference in the expression of HMGA1, P 0.01 between laryngeal carcinoma and the incisor margin of carcinoma (P < 0.01). Ezrin, (P0.01), one-way ANOVA analysis showed that HMGA1,Ezrin was related to histological grade, lymph node metastasis, clinical stage and lymph node metastasis, but was related to smoking, but correlated with the histological grade (P 0.05), lymph node metastasis (P < 0.05), clinical stage (P 0.05) and lymph node metastasis (P 0.05). There was no significant difference between age and sex (P0.05). (4). There was a positive correlation between the expression of HMGA1, Ezrin protein and mRNA (real time quantitative PCR:r=0.385,P=0.000;) in laryngeal carcinoma. Immunohistochemical staining: x2 / 7.172, r = 0.379, P = 0.012 / Western blot:r=0.412,P=0.000). Conclusion: (1) the expression of HMGA1, Ezrin and mRNA in laryngeal carcinoma is abnormal, which is higher than that in normal laryngeal epithelial tissue, and the positive rate of HMGA1,Ezrin expression gradually increases with the degree of malignant transformation of laryngeal dysplasia. (2) the expression of HMGA1, Ezrin and mRNA were positively correlated with lymph node metastasis, pathological grade and clinical stage, but not with age, sex or tumor location. (3) the increased expression of HMGA1 and Ezrin in laryngeal carcinoma may play a synergistic role in the carcinogenesis and development of laryngeal carcinoma, which can be used as biomarkers for predicting metastasis and predicting prognosis of laryngeal carcinoma.
