【摘要】：目的動態(tài)觀察大鼠角膜堿燒傷后Toll樣受體-2(Toll like-receptor 2,TLR-2)、Toll樣受體-4(Toll like-receptor 4,TLR-4)與炎性因子白細(xì)胞介素-1β(interleukin-1,IL-1β)、腫瘤壞死因子-α(tumor necrosis factor-α,TNF-α)的表達(dá)情況及變化規(guī)律,分析Toll樣受體家族(Toll like-receptor familys,TLRs)與炎性因子IL-1β、TNF-α的表達(dá)是否存在相關(guān)關(guān)系。方法選用健康SPF級SD大鼠40只,右眼為燒傷實(shí)驗(yàn)眼,左眼為正常對照眼,用濃度為1 mol·L-1Na OH建立Ⅲ級角膜堿燒傷模型(模型建立時(shí)及模型建立后出現(xiàn)大鼠角膜燒傷程度不是Ⅲ級、大鼠角膜穿孔或前房積血等情況,均予以剔除,最后隨機(jī)選取32只符合條件的大鼠進(jìn)行后續(xù)實(shí)驗(yàn)),隨機(jī)分為堿燒傷后3d組、7d組、14d組、21d組,每組8眼。分別在模型制作后觀察大鼠眼前節(jié),評估其角膜炎癥指數(shù),然后摘取大鼠眼球,行HE染色,計(jì)算炎癥細(xì)胞數(shù),同時(shí)用Western blot蛋白檢測法檢測大鼠角膜中TLR-2、TLR-4、IL-1β、TNF-α表達(dá)情況,分析各組差異,評估TLRs與炎性因子的相關(guān)性。結(jié)果在正常大鼠角膜中可檢測到TLR-2、TLR-4、IL-1β、TNF-α蛋白少量表達(dá),堿燒傷后其表達(dá)量增加,并于堿燒傷后7d達(dá)到峰值,以后逐漸遞減,各組間(3d組、7d組、14d組、21d組)比較差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05)。相關(guān)性分析:TLR-2與IL-1β(r=0.986,P0.05)、TNF-α(r=0.986,P0.05)的表達(dá)量呈正相關(guān),TLR-4與IL-1β(r=0.975,P0.05)、TNF-α(r=0.990,P0.05)的表達(dá)量亦呈正相關(guān)。結(jié)論TLRs參與并可能啟動角膜堿燒傷的免疫炎癥反應(yīng),介導(dǎo)炎性因子的產(chǎn)生。
