維生素D受體及其基因多態(tài)性與鼻咽癌的關(guān)聯(lián)

【摘要】:目的:探討維生素D受體(VDR)及其基因多態(tài)性與鼻咽癌的關(guān)聯(lián)。方法:采用熒光定量PCR檢測48例鼻咽癌患者(鼻咽癌組)和48例對照者(對照組)外周血單個核細胞中的VDR mRNA。運用TaqMan-MGB熒光探針PCR法檢測所有研究對象的FokI和ApaI位點基因型。結(jié)果:鼻咽癌組VDR mRNA△Ct值為9.32±0.91,對照組為7.31±1.04,鼻咽癌組VDR mRNA表達量顯著低于對照組(t=10.08,P0.01)。采用多因素Logistic回歸分析,結(jié)果顯示FokI點Ff基因型在鼻咽癌組中的分布顯著高于對照組(調(diào)整OR=1.97,95%CI=1.33~2.91)。未發(fā)現(xiàn)ApaI位點各基因型在對照組和鼻咽癌組中分布存在差異。進一步采用單倍型分析,結(jié)果顯示,與fa單倍型相比,Fa單倍型在對照組中的分布顯著高于鼻咽癌組(調(diào)整OR=0.65,95%CI=0.48~0.87)。2組FokI位點不同基因型中VDR mRNA表達水平差異有統(tǒng)計學意義(F=194.5,P0.01)。結(jié)論:VDR基因遺傳變異與鼻咽癌有密切關(guān)聯(lián)。
[Abstract]:Objective: to investigate the association of vitamin D receptor (VDR) and its gene polymorphism with nasopharyngeal carcinoma. Methods: VDR mRNA. in peripheral blood mononuclear cells of 48 patients with nasopharyngeal carcinoma (NPC group) and 48 controls (control group) was detected by fluorescence quantitative PCR. The genotypes of FokI and ApaI loci in all subjects were detected by TaqMan-MGB fluorescence probe PCR method. Results: VDR mRNA Ct was 9.32 鹵0.91in nasopharyngeal carcinoma group and 7.31 鹵1.04in control group. The expression of VDR mRNA in nasopharyngeal carcinoma group was significantly lower than that in control group (t = 10.08, P0.01). Multivariate Logistic regression analysis showed that the distribution of FokI point Ff genotype in nasopharyngeal carcinoma was significantly higher than that in control group (adjusted OR=1.97,95%CI=1.33~2.91). There was no significant difference in the distribution of ApaI locus genotypes between control group and nasopharyngeal carcinoma group. The results of haplotype analysis showed that the distribution of Fa haplotype in control group was significantly higher than that in nasopharyngeal carcinoma group (adjusted OR=0.65,95%CI=0.48~0.87). There were significant differences in VDR mRNA expression in different genotypes of FokI locus between two groups (FF194.5, P0.01). Conclusion: the genetic variation of VDR gene is closely related to nasopharyngeal carcinoma.
