【摘要】：目的：探討槲皮素對(duì)體外培養(yǎng)人翼狀胬肉成纖維細(xì)胞(HPF)增殖作用的影響，尋找預(yù)防翼狀胬肉的發(fā)生和減少翼狀胬肉復(fù)發(fā)的新方法。 方法：1、用0～160mg/L槲皮素作用體外培養(yǎng)的HPF，24、48、72小時(shí)后CCK-8法檢測(cè)藥物對(duì)HPF的影響。2、Hoechst33342染色法：正常細(xì)胞攝取Hoechst33342染料后呈現(xiàn)藍(lán)色熒光，而凋亡細(xì)胞攝取Hoechst33342能力比正細(xì)胞強(qiáng)，呈強(qiáng)藍(lán)色熒光。用0～160mg/L槲皮素作用體外培養(yǎng)的HPF，24小時(shí)后加入Hoechst33342染色，10分鐘后在熒光顯微鏡下觀察熒光強(qiáng)弱。 結(jié)果：1、在20～80mg/L范圍內(nèi)槲皮素作用24h后能顯著抑制HPF的增殖(P0.05)，，呈時(shí)間依賴性和濃度依賴性。在0～160mg/L范圍內(nèi)槲皮素作用48h、72h后能顯著抑制HPF的增殖(P0.05)，呈時(shí)間依賴性和濃度依賴性。2、Hoechst33258染色法顯示在10～160mg/L范圍內(nèi)槲皮素作用24小時(shí)后，隨槲皮素劑量的增加細(xì)胞凋亡增加。 結(jié)論：槲皮素可顯著抑制HPF的增殖和促細(xì)胞凋亡。
[Abstract]:Objective: to investigate the effect of quercetin on the proliferation of human pterygium fibroblasts (HPF) in vitro and to find a new method to prevent the occurrence of pterygium and reduce the recurrence of pterygium. Methods: 1. CCK-8 method was used to detect the effect of 0~160mg/L quercetin on HPF after HPF,24,48,72 hours. 2Hoechst33342 staining method: normal cells showed blue fluorescence after uptake of Hoechst33342 dye, but apoptotic cells had stronger ability of Hoechst33342 uptake than positive cells. Strong blue fluorescence. The HPF,24 cultured in vitro was treated with 0~160mg/L quercetin for hours and then Hoechst33342 staining was added. The fluorescence intensity was observed under fluorescence microscope for 10 minutes. Results: 1. Quercetin could significantly inhibit the proliferation of HPF in the range of 20~80mg/L for 24 h (P0.05), in a time-and concentration-dependent manner. The proliferation of HPF was significantly inhibited by quercetin in the range of 0~160mg/L for 48 h or 72 h (P0.05) in a time-and concentration-dependent manner. (2) Hoechst33258 staining showed that the apoptosis of HPF increased with the increase of quercetin dose in the range of 10~160mg/L for 24 hours. Conclusion: quercetin can significantly inhibit the proliferation and promote apoptosis of HPF.
