益生菌DNA干預(yù)對變應(yīng)性鼻炎療效的動物實驗研究

【摘要】:目的 益生菌DNA滴鼻干預(yù)對卵清蛋白(OVA)誘發(fā)的變應(yīng)性鼻炎療效的動物模型研究,探討小鼠鼻黏膜中調(diào)節(jié)性T細(xì)胞(Treg細(xì)胞)及血清Th1/ Th2細(xì)胞因子水平。 方法 培養(yǎng)益生菌,提取DNA,取6-8周Balb/c小鼠隨機(jī)分為益生菌DNA滴鼻組及生理鹽水對照組,每組15只。益生菌滴鼻組:30%的卵清蛋白(OVA)生理鹽水溶液,每毫升加入AL(OH)330 mg腹腔注射大鼠,隔日1次,每次1 ml,7次后用5%的OVA液滴鼻,每側(cè)l0ul,每天1次,共7次,致敏的同時取已提取的DNA液滴鼻,每側(cè)5ul(25ug/ml),每天一次。對照組:OVA造模方法同益生菌滴鼻組,但以生理鹽水代替益生菌DNA液滴鼻。采用ELISA法檢測各組血清IgE及IL-4,IL-10,IFN-γ,IFN-γ/ IL-4水平。分析鼻黏膜中Foxp3陽性細(xì)胞數(shù),并對鼻黏膜行組織形態(tài)學(xué)觀察。組間用t檢驗。 結(jié)果 1.益生菌DNA滴鼻組IgE顯著低于對照組(p0.05), 2.益生菌DNA滴鼻組IL-10,IFN-γ,IL-4均低于對照組(p0.05),而IFN-γ/IL-4高于對照組(p0.05)。 3.益生菌DNA滴鼻組鼻粘膜中Foxp-3陽性細(xì)胞數(shù)顯著高于對照組(p0.05)。 4.益生菌DNA滴鼻組鼻粘膜較對照組出現(xiàn)明顯的形態(tài)學(xué)改變。 結(jié)論 1.益生菌DNA滴鼻干預(yù)通過降低動物模型IgE水平,緩解Ⅰ型變態(tài)反應(yīng)性。 2.益生菌DNA滴鼻通過調(diào)整模型動物Th1/Th2細(xì)胞因子平衡,,抑制Th2細(xì)胞反應(yīng),對變應(yīng)性鼻炎起一定防治作用。 3.益生菌DNA滴鼻早期干預(yù)可提高動物模型鼻粘膜Foxp3陽性細(xì)胞數(shù)抑制小鼠AR的進(jìn)展。
[Abstract]:Objective to investigate the effects of probiotic DNA nasal drip on allergic rhinitis induced by ovalbumin (OVA) and to investigate the levels of regulatory T cells (Treg cells) and serum Th1/ Th2 cytokines in mouse nasal mucosa. Methods probiotics were cultured and Balb/c mice were randomly divided into probiotic DNA nasal dropping group and normal saline control group for 6 to 8 weeks with 15 mice in each group. Probiotic nasal drip group: 30% ovalbumin (OVA) saline solution was injected intraperitoneally into rats with AL (OH) 330 mg per milliliter. Once a second day, 1 ml,7 per day, 5% OVA solution was used on each side of the nose, 10% per side, once a day, for 7 times. At the same time, the extracted DNA solution was nasal drops, 5ul (25ug/ml) on each side, once a day. The control group was treated with probiotic nasal drip, but normal saline was used instead of probiotic DNA solution. Serum IgE and IL-4,IL-10,IFN- 緯 / IL-4 levels were detected by ELISA assay. The number of Foxp3 positive cells in nasal mucosa was analyzed and the histomorphology of nasal mucosa was observed. T test was used between groups. Result 1. The IgE of probiotic DNA nasal drip group was significantly lower than that of control group (p0.05). The levels of IL-10,IFN- 緯 -IL-4 in probiotic DNA nasal drip group were lower than those in control group (p0.05), while IFN- 緯 -IL-4 was higher than that in control group (p0.05). The number of Foxp-3 positive cells in nasal mucosa of probiotic DNA dropping group was significantly higher than that of control group (p0.05).
