IL-9、IL-10在慢性鼻竇炎性疾病中的表達(dá)及意義

【摘要】:目的 Th9細(xì)胞是新近發(fā)現(xiàn)的新型的功能性輔助性T細(xì)胞亞群,主要表達(dá)IL-9和IL-10兩種細(xì)胞因子。IL.9.IL-10與多種疾病的發(fā)生發(fā)展密切相關(guān),特別在呼吸系統(tǒng)炎性疾病中發(fā)揮重要作用。本文旨在觀察IL-9.IL-10在慢性鼻竇炎性疾病中的表達(dá)及其與局部嗜酸性粒細(xì)胞浸潤(rùn)的關(guān)系,進(jìn)一步探討其在慢性鼻竇炎性疾病中的可能作用機(jī)理。 方法 選取住院接受治療的慢性鼻-鼻竇炎(chronic rhinosinusitis,CRS)患者30例和真菌性鼻-鼻竇炎(fungal rhino-sinusitis,FRS)患者13例,常規(guī)行FESS手術(shù),術(shù)中收集病變鼻竇竇腔粘膜組織,并以18例鼻腔鼻竇非炎性疾病患者為對(duì)照組,收集其鼻腔非炎粘膜組織。采用H-E染色法觀察組織形態(tài)并檢測(cè)嗜酸性粒細(xì)胞數(shù);免疫組織化學(xué)S-P二步法檢查組織中IL-9、IL-10蛋白相對(duì)含量;半定量逆轉(zhuǎn)錄-聚合酶鏈?zhǔn)椒磻?yīng)(reverse transcriptase polymerase chain reaction,RT-PCR)檢測(cè)組織中IL-9.IL.10基因mRNA表達(dá)。所有資料使用SPSS13.0統(tǒng)計(jì)分析軟件進(jìn)行統(tǒng)計(jì)分析,采用方差分析及Pearson相關(guān)性分析方法,P0.05為差異有統(tǒng)計(jì)學(xué)意義。 結(jié)果 (1)慢性鼻-鼻竇炎組與真菌性鼻-鼻竇炎組患者病變鼻竇粘膜組織內(nèi)均見大量嗜酸性粒細(xì)胞浸潤(rùn),明顯高于對(duì)照組的鼻腔非炎粘膜組織,P0.05:(2)IL-9、IL-10蛋白在慢性鼻-鼻竇炎與真菌性鼻-鼻竇炎患者病變鼻竇粘膜組織內(nèi)均呈陽性表達(dá),且表達(dá)量明顯高于鼻腔非炎粘膜組織,相比差異有統(tǒng)計(jì)學(xué)意義P0.05;但在慢性鼻-鼻竇炎與真菌性鼻-鼻竇炎組之間差異無統(tǒng)計(jì)學(xué)意義,P0.05;(3)慢性鼻-鼻竇炎組與真菌性鼻-鼻竇炎組患者病變鼻竇粘膜內(nèi)IL-9基因和IL-10基因mRNA均有表達(dá),且其相對(duì)表達(dá)量均高于對(duì)照組,P0.05;但在慢性鼻-鼻竇炎組與真菌性鼻-鼻竇炎組之間差異無統(tǒng)計(jì)學(xué)意義,P0.05;(4)在慢性鼻-鼻竇炎患者病變鼻竇粘膜內(nèi)IL-9與IL-10蛋白相對(duì)表達(dá)量與局部嗜酸性粒細(xì)胞浸潤(rùn)數(shù)均存在明顯相關(guān)性P0.05。 結(jié)論 (1)慢性鼻-鼻竇炎與真菌性鼻-鼻竇炎患者病變鼻竇粘膜內(nèi)可見大量嗜酸性粒細(xì)胞浸潤(rùn),為其重要病理性特征之一;(2)IL-9與IL-10在慢性鼻-鼻竇炎與真菌性鼻-鼻竇炎患者病變鼻竇粘膜均有高表達(dá),二者可能參與鼻竇炎性疾病的發(fā)生發(fā)展;(3)IL-9、IL-10與局部嗜酸性粒細(xì)胞浸潤(rùn)密切相關(guān),其可能通過嗜酸性粒細(xì)胞影響鼻竇炎性疾病的發(fā)生發(fā)展。
[Abstract]:Objective Th9 cells are a new type of functional helper T cell subsets. The expression of IL-9 and IL-10 cytokines, IL.9.IL-10, is closely related to the occurrence and development of many diseases. Especially in the respiratory inflammatory diseases play an important role. The purpose of this study was to investigate the expression of IL-9.IL-10 in chronic sinusitis and its relationship with local eosinophil infiltration, and to explore its possible mechanism in chronic sinusitis. Methods 30 patients with chronic rhinosinusitis and 13 patients with fungal rhino-sinusitis were selected for FESS operation. The non-inflammatory mucosa of nasal cavity and sinuses were collected from 18 patients with non-inflammatory diseases of nasal cavity and paranasal sinuses as control group. Tissue morphology was observed by H-E staining and the number of eosinophils was detected by immunohistochemistry S-P two-step method was used to detect the relative content of IL-9 and IL-10 protein in tissues. Semi-quantitative reverse transcription-polymerase chain reaction (reverse transcriptase polymerase chain RT-PCR) was used to detect the expression of IL-9.IL.10 gene mRNA in tissues. All the data were statistically analyzed by SPSS13.0 statistical analysis software. The difference was statistically significant by means of ANOVA and Pearson correlation analysis (P0.05). Results (1) in both chronic rhinosinusitis group and fungal rhinosinusitis group, a large number of eosinophils infiltrated in the mucosa of the nasal sinuses. The expression of IL-9 IL-10 protein was significantly higher in patients with chronic rhinosinusitis and fungal rhinosinusitis, and the expression of IL-9 IL-10 protein was significantly higher than that in non-inflammatory nasal mucosa tissue, and the expression of IL-9 IL-10 protein was significantly higher in patients with chronic rhinosinusitis and fungal rhinosinusitis. But there was no significant difference between chronic rhinosinusitis and fungal rhinosinusitis. (3) the expression of IL-9 gene and IL-10 gene mRNA in nasal mucosa of patients with chronic rhinosinusitis and fungal rhinosinusitis were higher than that of control group (P 0.05). But there was no significant difference between chronic rhinosinusitis group and fungal rhinosinusitis group (P 0.05). (4) in patients with chronic rhinosinusitis, there was a significant correlation between the relative expression of IL-9 and IL-10 protein and the number of eosinophil infiltration (P0.05). Conclusion (1) in patients with chronic rhinosinusitis and fungal rhinosinusitis, a large number of eosinophils infiltration can be seen in the mucosa of the nasal sinuses, which is one of the important pathological features. (2) IL-9 and IL-10 were highly expressed in the nasal mucosa of patients with chronic rhinosinusitis and fungal rhinosinusitis, which may be involved in the occurrence and development of sinusitis, (3) IL-9 IL-10 was closely related to local eosinophils infiltration. It may affect the development of sinusitis disease through eosinophil.
