【摘要】：目的構(gòu)建高度近視豚鼠脈絡(luò)膜血管新生(choroidal neovascularization,CNV)模型,探討基質(zhì)金屬蛋白酶(matrix metalloproteinase-2,MMP-2)和基質(zhì)金屬蛋白酶抑制劑(tissue inhibitor-2 of metalloproteinase,TIMP-2)在高度近視CNV形成中的作用。方法 72只2周齡三色豚鼠隨機(jī)分為實(shí)驗(yàn)組(n=36)和對(duì)照組(n=36),實(shí)驗(yàn)組右眼行6周形覺(jué)剝奪誘導(dǎo)高度近視。兩組各選取30只豚鼠右眼行532 nm激光光凝視網(wǎng)膜誘導(dǎo)CNV。于激光前和光凝后7、14、21、28、35天進(jìn)行免疫組化觀察光凝區(qū)域MMP-2和TIMP-2的表達(dá),real-time PCR檢測(cè)視網(wǎng)膜色素上皮層(retinal pigment epithelium,RPE)-脈絡(luò)膜-鞏膜復(fù)合物中的MMP-2和TIMP-2mRNA表達(dá)對(duì)MMP-2和TIMP-2免疫組化陽(yáng)性表達(dá)的積分光密度值(integral optical desnsity,IDO)及mRNA相對(duì)表達(dá)水平進(jìn)行統(tǒng)計(jì)學(xué)分析。結(jié)果激光光凝后實(shí)驗(yàn)組和對(duì)照組MMP-2和TIMP-2表達(dá)均明顯上調(diào),MMP-2于光凝后21天、TIMP-2于光凝后28天時(shí)表達(dá)高峰。激光前及光凝后各觀察時(shí)間點(diǎn),實(shí)驗(yàn)組MMP-2陽(yáng)性表達(dá)和mRNA相對(duì)表達(dá)量均明顯高于對(duì)照組,差異有統(tǒng)計(jì)學(xué)意義(P0.05);而激光前實(shí)驗(yàn)組TIMP-2表達(dá)較對(duì)照組明顯減少(P0.05),光凝后各時(shí)間點(diǎn)兩組TIMP-2表達(dá)差異無(wú)統(tǒng)計(jì)學(xué)意義。結(jié)論MMP-2和TIMP-2與高度近視CNV形成密切相關(guān),MMP-2與TIMP-2表達(dá)平衡紊亂可能參與高度近視CNV的發(fā)生發(fā)展。
[Abstract]:Objective to study the role of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor-2 of metalloproteinase (TIMP-2) in the formation of high myopia (CNV) in guinea pigs with high myopia by establishing a choroidal neovascularization model. Methods 72 two-week-old tricolor guinea pigs were randomly divided into two groups: the experimental group (n = 36) and the control group (n = 36). The right eye of the experimental group was induced by 6 weeks of form deprivation to induce high myopia. 30 guinea pigs in each group were treated with 532 nm laser photocoagulation to induce CNV. Immunohistochemical observation of MMP-2 and TIMP-2 expression in the photocoagulation region before and after photocoagulation: detection of MMP-2 and TIMP-2mRNA expression in (retinal pigment epithelium of retinal pigment epithelium layer by real-time PCR and MMP-2 and TIMP-2mRNA expression in chorio-scleral complexes positive for MMP-2 and TIMP-2 The integrated optical density (integral optical) and the relative expression level of mRNA were analyzed statistically. Results after laser photocoagulation, the expression of MMP-2 and TIMP-2 in the experimental group and the control group were significantly up-regulated, and the expression of TIMP-2 was significantly increased on the 21st day after photocoagulation, and the peak expression of TIMP-2 was observed on the 28th day after photocoagulation. The positive expression of MMP-2 and the relative expression of mRNA in the experimental group were significantly higher than those in the control group before and after photocoagulation. The difference was statistically significant (P0.05), while the expression of TIMP-2 in the experimental group was significantly lower than that in the control group (P0.05). There was no significant difference in the expression of TIMP-2 between the two groups at each time point after photocoagulation. Conclusion MMP-2 and TIMP-2 are closely related to the formation of CNV in high myopia. The imbalance of MMP-2 and TIMP-2 expression may be involved in the occurrence and development of CNV in high myopia.
