ALOX15在慢性鼻竇炎伴鼻息肉中表達(dá)及糖皮質(zhì)激素對(duì)其作用研究

【摘要】:目的研究花生四烯酸酯15-脂氧合酶(arachidonate 15-lipoxygenase,ALOX15)在嗜酸粒細(xì)胞型慢性鼻竇炎伴鼻息肉(eosinophilic chronic rhinosinusitis with nasal polyps,ECRSw NP)及非嗜酸粒細(xì)胞型慢性鼻竇炎伴鼻息肉(nonesinophilc chronic rhinosinusitis with nasal polyps,non ECRSw NP)中表達(dá),并研究口服糖皮質(zhì)激素對(duì)其表達(dá)的影響。方法所有標(biāo)本均由首都醫(yī)科大學(xué)附屬北京同仁醫(yī)院標(biāo)本庫中獲得,采用real-time PCR及免疫組織化學(xué)染色研究ALOX15表達(dá)與蛋白定位。比較激素治療前ECRSw NP、non ECRSw NP及健康對(duì)照組中ALOX15 mRNA表達(dá)水平、蛋白定位及表達(dá),并比較同一病人激素治療前后mRNA及蛋白表達(dá)分布差異。結(jié)果 ECRSw NP組中ALOX15 mRNA及蛋白表達(dá)量高于健康對(duì)照組及non ECRSw NP組,差異有統(tǒng)計(jì)學(xué)意義(均P=0.000)。健康對(duì)照組、non ECRSw NP組間差異無統(tǒng)計(jì)學(xué)意義(P=0.594,P=0.550)。ALOX15蛋白表達(dá)于上皮細(xì)胞、漿細(xì)胞、中性粒細(xì)胞及嗜酸性粒細(xì)胞等多種細(xì)胞。ECRSw NP組激素治療前后ALOX15 mRNA和蛋白表達(dá)差異有統(tǒng)計(jì)學(xué)意義(P=0.004,P=0.024)。結(jié)論 ALOX15在ECRSw NP中表達(dá)顯著高于non ECRSw NP及健康對(duì)照組,口服糖皮質(zhì)激素1周能抑制其表達(dá)。
[Abstract]:Objective to investigate the expression of arachidonate 15-lipoxygenase (ALOX15) in chronic eosinophil sinusitis with (eosinophilic chronic rhinosinusitis with nasal polypsm ECRSw NP and non-eosinophil chronic sinusitis with (nonesinophilc chronic rhinosinusitis with nasal polypsnon ECRSw NP). And to study the effect of oral glucocorticoid on its expression. Methods all specimens were collected from the specimen bank of Tongren Hospital affiliated to Capital Medical University. ALOX15 expression and protein localization were studied by real-time PCR and immunohistochemical staining. The expression of ALOX15 mRNA, protein localization and protein expression in ECRSw NPnon ECRSw NP and healthy control group were compared before and after hormone therapy, and the differences of mRNA and protein expression before and after hormone therapy in the same patient were compared. Results the expression of ALOX15 mRNA and protein in ECRSw NP group was significantly higher than that in healthy control group and non ECRSw NP group (P < 0. 000). The expression of ALOX15 protein in epithelial cells, plasma cells, neutrophils and eosinophils was not significantly different between the control group and control group (P0. 594P0. 550). The expression of ALOX15 mRNA and protein in ECRSw NP group was significantly different before and after hormone treatment (P0. 004, P0. 024). Conclusion the expression of ALOX15 in ECRSw NP is significantly higher than that in non ECRSw NP and healthy control group. Glucocorticoid can inhibit the expression of ALOX15 in ECRSw NP for 1 week.
