漢黃芩素體外抗人喉癌Hep2細(xì)胞增殖、凋亡和侵襲的作用及機(jī)制研究

【摘要】:目的研究漢黃芩素對人喉癌Hep-2細(xì)胞增殖、凋亡及侵襲能力的影響,并探討其作用機(jī)制。方法運用CCK-8檢測漢黃芩素對人喉癌Hep-2細(xì)胞增殖活性的影響。運用流式細(xì)胞儀檢測漢黃芩素對人喉癌Hep-2細(xì)胞凋亡的影響。運用Hoechst染色法檢測漢黃芩素對人喉癌Hep-2細(xì)胞核形態(tài)變化的影響。運用transwell侵襲實驗比較漢黃芩素對人喉癌Hep-2細(xì)胞侵襲能力的影響。運用Western blot檢測漢黃芩素對人喉癌Hep-2細(xì)胞h TERT、Bcl-2、Bax蛋白表達(dá)水平的影響。結(jié)果 CCK-8法發(fā)現(xiàn)漢黃芩素能抑制人喉癌Hep-2細(xì)胞增殖,其作用呈濃度依賴性。流式細(xì)胞術(shù)分析發(fā)現(xiàn)漢黃芩素能誘導(dǎo)人喉癌Hep-2細(xì)胞發(fā)生凋亡。Hoechst染色發(fā)現(xiàn)漢黃芩素能誘導(dǎo)人喉癌Hep-2細(xì)胞核形態(tài)變化。Transwell實驗發(fā)現(xiàn)漢黃芩素能抑制人喉癌Hep-2細(xì)胞的體外侵襲能力。Western blot檢測發(fā)現(xiàn)測漢黃芩素能有效降低人喉癌Hep-2細(xì)胞h TERT、Bcl-2蛋白表達(dá)水平以及增加Bax蛋白表達(dá)水平(P0.05)。結(jié)論漢黃芩素在體外能夠有效抑制人喉癌Hep-2細(xì)胞增殖,誘導(dǎo)細(xì)胞凋亡,同時抑制喉癌細(xì)胞的體外侵襲能力。其作用可能與調(diào)控Bcl-2蛋白家族及h TERT蛋白表達(dá)水平相關(guān)。
[Abstract]:Objective to study the effects of baicalin on proliferation, apoptosis and invasion of human laryngeal carcinoma Hep-2 cells and its mechanism. Methods the effect of baicalin on the proliferation of human laryngeal carcinoma Hep-2 cells was detected by CCK-8. The effect of baicalin on apoptosis of human laryngeal carcinoma Hep-2 cells was detected by flow cytometry. The effects of baicalin on the nuclear morphologic changes of Hep-2 in human laryngeal carcinoma were detected by Hoechst staining. Transwell invasion assay was used to compare the effect of baicalin on the invasion ability of human laryngeal carcinoma Hep-2 cells. The effect of baicalin on the expression of hTERTnBcl-2mBax protein in human laryngeal carcinoma Hep-2 cells was detected by Western blot. Results CCK-8 assay showed that baicalin could inhibit the proliferation of human laryngeal carcinoma Hep-2 cells in a concentration dependent manner. Flow cytometric analysis showed that baicalin could induce apoptosis of human laryngeal carcinoma Hep-2 cells. Hoechst staining found that Chinese baicalin could induce nuclear morphologic changes of human laryngeal carcinoma Hep-2. Transwell assay found that Chinese baicalin could inhibit human laryngeal cancer Hep-2 cells in vitro. Western blot assay showed that baicalin could effectively decrease the expression of hTERTnBcl-2 protein and increase the expression of Bax protein in human laryngeal carcinoma Hep-2 cells (P0.05). Conclusion baicalin can effectively inhibit the proliferation and apoptosis of human laryngeal carcinoma Hep-2 cells in vitro and inhibit the invasiveness of human laryngeal carcinoma cells in vitro. Its role may be related to the regulation of Bcl-2 protein family and the expression of h TERT protein.
