硫酸鋅對紫外線誘導的大鼠晶狀體氧化損傷的保護作用

【摘要】:目的 探討硫酸鋅(ZnSO_4)對紫外線誘導的大鼠晶狀體氧化損傷的保護作用。 方法 培養(yǎng)離體大鼠晶狀體,,將其隨機分為三組:(1)陰性對照組(正常組);(2)陽性對照組(紫外線照射組);(3)實驗組(紫外線照射+ZnSO_4組)。觀察晶狀體的混濁程度,取各組晶狀體標本進行蘇木素/伊紅染色(HE)染色,光鏡下觀察組織形態(tài)學變化,檢測晶狀體組織中谷胱甘肽過氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)及晶狀體中熱休克蛋白70(HSP70)表達的變化。 結果 陰性對照組各時段的晶狀體基本保持透明;陽性對照組晶狀體隨著培養(yǎng)時間的延長逐漸混濁;實驗組晶狀體隨著培養(yǎng)時間的延長逐漸混濁,但與同時段陽性對照組相比,混濁程度減輕。光學顯微鏡下見陰性對照組晶狀體組織結構基本正常;陽性對照組與實驗組晶狀體組織結構有不同程度的損傷;實驗組晶狀體損傷程度較同時段陽性對照組減輕。陽性對照組晶狀體中GSH-Px和SOD活性低于同時段陰性對照組,差異有統(tǒng)計學意義(P0.05);實驗組晶狀體中GSH-Px和SOD活性高于同時段陽性對照組,差異有統(tǒng)計學意義(P0.05)。陽性對照組晶狀體中HSP70的表達與同時段陰性對照組相比,表達增加,差異有統(tǒng)計學意義(P0.05);實驗組晶狀體中HSP70的表達與同時段陽性對照組相比,表達增加,差異有統(tǒng)計學意義(P0.05)。 結論 ZnSO_4可以誘導離體大鼠晶狀體中HSP70的表達,可以提高氧化損傷晶狀體的抗氧化能力,在維持晶狀體透明性方面起到了一定的作用。
[Abstract]:Objective to investigate the protective effect of zinc sulfate (ZnSO _ 4) on ultraviolet induced oxidative injury of rat lens. Methods isolated rat lens were cultured and randomly divided into three groups: (1) negative control group (normal group,); (_ 2), positive control group (ultraviolet radiation group,); (_ 3), experimental group (ultraviolet radiation); (_ 3 group). The degree of opacity of lens was observed. Hematoxylin / eosin (HE) staining and histomorphology were observed under light microscope. The changes of glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) and heat shock protein 70 (HSP70) in lens were detected. Results the lens of the negative control group was basically transparent at every time, the lens of the positive control group was gradually opacified with the extension of culture time, and the lens of the experimental group was gradually opacified with the extension of the culture time. But compared with the positive control group at the same time, the degree of turbidity decreased. Under the optical microscope, the structure of the lens in the negative control group was basically normal, that in the positive control group and the experimental group was different degrees, and the degree of lens injury in the experimental group was less than that in the positive control group in the same period. The activities of GSH-Px and SOD in lens of positive control group were lower than those of negative control group in the same period (P0.05); the activities of GSH-Px and SOD in lens of experimental group were higher than that of positive control group in the same period (P0.05). The expression of HSP70 in the lens of the positive control group was higher than that of the negative control group in the same period (P0.05), while the expression of HSP70 in the lens of the experimental group was higher than that of the positive control group in the same period (P0.05). Conclusion ZnSOS _ 4 can induce the expression of HSP70 in isolated rat lens, increase the anti-oxidation ability of the injured lens, and play a certain role in maintaining the transparency of the lens.
