抗抑郁藥地昔帕明對變應(yīng)性鼻炎小鼠的緩解作用

【摘要】:背景:變應(yīng)性鼻炎是由IgE介導(dǎo)的超敏反應(yīng)。它是由于鼻粘膜對某些刺激因子過度敏感而產(chǎn)生超出生理范圍的鼻粘膜炎癥反應(yīng)。地昔帕明屬于三環(huán)類抗抑郁藥,同時有報道指出它對酸性鞘磷脂酶具有抑制作用。酸性鞘磷脂酶在炎癥反應(yīng)過程中發(fā)揮著必不可少的作用。地昔帕明對變應(yīng)性鼻炎小鼠的作用效果尚未有相關(guān)的報道。因此,本實驗的研究目的為,抗抑郁藥地昔帕明對變應(yīng)性鼻炎小鼠的鼻部癥狀是否療效。 方法:雄性BALB/C小鼠用卵清蛋白和氫氧化鋁混合液腹腔注射致敏,激發(fā)階段用卵清蛋白滴鼻。變應(yīng)性鼻炎模型造模成功后,每次卵清蛋白滴鼻后,地昔帕明灌胃。觀察變應(yīng)性鼻炎小鼠的行為學(xué),包括噴嚏和抓鼻次數(shù),來判患病小鼠的嚴(yán)重程度。通過ELISA的方法檢測血清卵清蛋白特異性IgE及鼻腔灌洗液中細(xì)胞因子IL-4和INF-γ的水平。并且應(yīng)用流式細(xì)胞儀來檢測小鼠脾細(xì)胞中調(diào)節(jié)性T細(xì)胞和Th17細(xì)胞來探討相關(guān)的作用機制。 結(jié)果:反復(fù)用地昔帕明灌胃可以減輕受激發(fā)小鼠過敏癥狀(噴嚏及抓鼻次數(shù))的發(fā)生,并且與地昔帕明的計量呈相關(guān)性。它還可以減少血清中卵清蛋白特異性IgE的水平和鼻腔灌洗液中IL-4的水平。盡管如此,地昔帕明對鼻腔灌洗液中INF-γ的水平?jīng)]有明顯的作用。此外,地昔帕明還可以通過對Treg細(xì)胞和Th17細(xì)胞進(jìn)行調(diào)節(jié)來削弱變應(yīng)性鼻炎的氣道炎癥反應(yīng)。 結(jié)論:地昔帕明可以通過抑制產(chǎn)生血清卵清蛋白特異性IgE及IL-4,,并且調(diào)節(jié)脾細(xì)胞中CD4+CD25+Foxp3+T細(xì)胞和CD4+Th17細(xì)胞的比例來發(fā)揮其抗過敏的功效。在變應(yīng)性鼻炎的發(fā)病機制中,除已知的Th1/Th2平衡外,還存在Treg/Th17平衡,這為我們研究變應(yīng)性鼻炎免疫學(xué)機制提供了一條新的理論依據(jù),為以后變應(yīng)性鼻炎的治療提供了新思路。
[Abstract]:Background: allergic rhinitis is a hypersensitive response mediated by IgE. It is due to nasal mucosal hypersensitivity to certain irritants and the production of physiological range of nasal inflammation. Despermine is a tricyclic antidepressant and has been reported to have inhibitory effects on acid sphingolipase. Acid sphingolipase plays an essential role in inflammatory reaction. There is no report on the effect of desparmine on allergic rhinitis in mice. Therefore, the aim of this study was to determine the efficacy of the antidepressant Dexiparam on nasal symptoms in allergic rhinitis mice. Methods: male BALB / C mice were sensitized by intraperitoneal injection of ovalbumin and aluminum hydroxide. After the model of allergic rhinitis was established successfully, after ovalbumin drops, desicamine was administered to the stomach. Observe the behavior of allergic rhinitis mice, including sneezing and nasal scratching times, to determine the severity of the disease in mice. The levels of serum ovalbumin specific IgE and cytokines IL-4 and INF- 緯 in nasal lavage fluid were detected by Elisa. Flow cytometry was used to detect regulatory T cells and Th17 cells in mouse spleen cells. Results: repeated oral administration of dixipramine reduced the incidence of allergic symptoms (sneezing and nasal scratching) in stimulated mice, and was correlated with the metrology of desipramine. It also reduced the serum level of ovalbumin specific IgE and the level of IL-4 in nasal lavage fluid. Nevertheless, desipramine had no significant effect on the level of INF- 緯 in nasal lavage fluid. In addition, despermine can weaken the airway inflammation of allergic rhinitis by regulating Treg cells and Th17 cells. Conclusion: Despermine can play its antiallergic effect by inhibiting the production of serum ovalbumin specific IgE and IL-4, and regulating the ratio of CD4 CD25 Foxp3 T cells to CD4 Th17 cells in spleen cells. In the pathogenesis of allergic rhinitis, in addition to the known Th1 / Th2 balance, there is Treg-Th17 balance, which provides a new theoretical basis for us to study the immune mechanism of allergic rhinitis, and provides a new idea for the treatment of allergic rhinitis.
