CR2、MLXIP、ZNF608和CSF1R在鼻咽癌組織中的表達(dá)及其臨床意義

【摘要】:背景和目的:在前期的研究中,我們通過利用能夠檢測(cè)14112個(gè)人類基因的cDNA芯片,對(duì)20例鼻咽癌患者組織樣本與15例鼻咽慢性炎癥患者組織樣本進(jìn)行表達(dá)譜分析比較,找出在鼻咽癌組織中有明顯差異表達(dá)的基因。本實(shí)驗(yàn)的研究目的是:1、從中選出鼻咽癌相關(guān)基因補(bǔ)體受體2(CR2)、MLX相互作用的蛋白(MLXIP),在50例鼻咽癌組織中進(jìn)行RT-PCR,驗(yàn)證其是否與基因芯片表達(dá)結(jié)果相一致;2、從中選出放射敏感相關(guān)基因鋅指蛋白608(ZNF608)、集落刺激因子1受體(CSF1R),在17例鼻咽癌組織中進(jìn)行RT-PCR,驗(yàn)證其是否與基因芯片表達(dá)結(jié)果相一致;3、分析CR2、MLXIP、ZNF608和CSF1R這四個(gè)基因在鼻咽癌組織中的表達(dá)與臨床的相關(guān)性。 方法:實(shí)驗(yàn)分別對(duì)50例、17例低分化鼻咽癌組織進(jìn)行熒光實(shí)時(shí)定量RT-PCR,驗(yàn)證CR2、MLXIP、ZNF608和CSF1R在鼻咽癌組織中的表達(dá),并將其與臨床進(jìn)行相關(guān)性分析。 結(jié)果: 1、我們選用CR2和MXLIP在50例低分化鼻咽癌組織中進(jìn)行RT-PCR驗(yàn)證,通過另外3例鼻咽慢性炎癥組織的表達(dá)譜比較,RT-PCR結(jié)果顯示,CR2在41例(82%)鼻咽癌組織中表達(dá)下調(diào),MLXIP在42例(84%)鼻咽癌組織中表達(dá)上調(diào)。我們?cè)谇捌诘难芯恐幸呀?jīng)知道表達(dá)譜芯片的分析結(jié)果為,9個(gè)基因(q0.01)至少在17例(85%)鼻咽癌組織中有2倍差異表達(dá),其中8個(gè)下調(diào)基因(TAOK3,SLC16A2,PRB4,AMY2B,B3GALT4,MSMB,RPS27,CR2),1個(gè)上調(diào)基因(MXLIP)。所以RT-PCR結(jié)果與芯片表達(dá)結(jié)果相同。 2、鼻咽癌組織中,CR2和MLXIP的表達(dá)與患者年齡、性別、臨床分期、T分期及頸部淋巴結(jié)轉(zhuǎn)移情況均無關(guān)(P0.05)。 3、本次研究結(jié)果顯示ZNF608和CSF1R在鼻咽癌放射抗拒型組中表達(dá)上調(diào),在鼻咽癌放射敏感型組中表達(dá)下調(diào)。在前期的研究中,ZNF608和CSF1R這兩個(gè)基因在芯片結(jié)果中顯示為,在鼻咽癌放射抗拒型病例組中表達(dá)上調(diào),在鼻咽癌放射敏感型組中表達(dá)下調(diào),它們的表達(dá)差異倍數(shù)分別是4.10倍和1.89倍。表明RT-PCR驗(yàn)證結(jié)果與芯片表達(dá)結(jié)果一致。 4、鼻咽癌組織中,ZNF608和CSF1R的表達(dá)與患者年齡、臨床分期、T分期及頸部淋巴結(jié)轉(zhuǎn)移情況均無關(guān)(P0.05)。 結(jié)論: 1、與炎癥組織相比較,CR2在41例(82%)鼻咽癌組織中表達(dá)下調(diào),MXLIP在42例(84%)鼻咽癌組織中表達(dá)上調(diào),與芯片表達(dá)結(jié)果一致,,CR2和MLXIP在鼻咽癌組織中有明顯的差異表達(dá),表明CR2和MLXIP在低分化鼻咽癌的診斷上有一定的作用。 2、鼻咽癌組織中,CR2和MLXIP的表達(dá)與患者年齡、性別、臨床分期、T分期及頸部淋巴結(jié)轉(zhuǎn)移情況無相關(guān)性。 3、ZNF608和CSF1R在鼻咽癌放射抗拒型病例組中表達(dá)上調(diào),在鼻咽癌放射敏感型組中表達(dá)下調(diào),與芯片表達(dá)結(jié)果一致,ZNF608和CSF1R有望作為預(yù)測(cè)鼻咽癌放射敏感性的指標(biāo)。 4、鼻咽癌組織中,ZNF608和CSF1R的表達(dá)與患者年齡、臨床分期、T分期及頸部淋巴結(jié)轉(zhuǎn)移情況無相關(guān)性。
[Abstract]:Background & AIM: in previous studies, we compared the expression profiles of 20 nasopharyngeal carcinoma tissue samples with 15 nasopharyngeal chronic inflammation tissue samples by using cDNA microarrays capable of detecting 14112 human genes. To identify the genes that are differentially expressed in nasopharyngeal carcinoma (NPC) tissues. The purpose of this study was to select a complement receptor 2 (CR2) / MLX interacting protein (MLXIP) from 1 to 50 NPC tissues to verify whether it was consistent with the results of gene chip expression. 2. The radiosensitivity genes zinc finger protein 608 (ZNF608) and colony stimulating factor 1 receptor (CSF1R) were selected. RT-PCR was performed in 17 cases of nasopharyngeal carcinoma to verify whether the results were consistent with the results of gene chip expression. 3. To analyze the correlation between the expression of CR2 gene MLXIPP ZNF608 and CSF1R in nasopharyngeal carcinoma (NPC). Methods: the expression of CR2MLXIPNZNF608 and CSF1R in 50 cases of poorly differentiated nasopharyngeal carcinoma (NPC) and 17 cases of poorly differentiated nasopharyngeal carcinoma (NPC) were examined by real-time quantitative RT-PCR, and the correlation between CR2MLXIPNZNF608 and CSF1R was analyzed. Results: 1. CR2 and MXLIP were used for RT-PCR in 50 cases of poorly differentiated nasopharyngeal carcinoma. The results of RT-PCR showed that the expression of CR2 was down-regulated in 41 cases (82%) of nasopharyngeal carcinoma tissues, and the expression of MLXIP was up-regulated in 42 cases (84%) of nasopharyngeal carcinoma tissues. We have known in previous studies that 9 genes (q0.01) were twice differentially expressed in at least 17 cases (85%) of nasopharyngeal carcinoma (NPC), including 8 down-regulated genes (TAOK3, SLC16A2, PRB4, AMY2GALT4MMBRPS27CR2) and 1 up-regulated gene (MXLIP). So the results of RT-PCR and microarray expression were the same. 2. The expression of CR2 and MLXIP in nasopharyngeal carcinoma tissues was related to the age and sex of patients. Clinical staging T stage and cervical lymph node metastasis were not significant (P0.05). The results showed that the expression of ZNF608 and CSF1R was up-regulated in the radioresistance group of nasopharyngeal carcinoma and down-regulated in the radiosensitive group of nasopharyngeal carcinoma. In previous studies, two genes, ZNF608 and CSF1R, were found to be up-regulated in patients with nasopharyngeal carcinoma (NPC) and down-regulated in radiosensitive nasopharyngeal carcinoma (RRS). The differences were 4.10 and 1.89 times, respectively. The results of RT-PCR were consistent with the results of microarray expression. 4. The expression of ZNF608 and CSF1R in nasopharyngeal carcinoma was not correlated with age, clinical staging and cervical lymph node metastasis (P0.05). Conclusion: 1. Compared with inflammatory tissue, the expression of CR2 in 41 cases (82%) of nasopharyngeal carcinoma was down-regulated in 42 cases (84%) of nasopharyngeal carcinoma, which was consistent with the results of microarray expression. There was significant difference between CR2 and MLXIP in nasopharyngeal carcinoma. CR2 and MLXIP may play an important role in the diagnosis of poorly differentiated nasopharyngeal carcinoma. 2. The expression of CR2 and MLXIP in nasopharyngeal carcinoma is associated with age and sex. There was no correlation between clinical staging and cervical lymph node metastasis. 3The expression of ZNF608 and CSF1R was up-regulated in patients with radioresistance of nasopharyngeal carcinoma, but down-regulated in radiosensitive type of nasopharyngeal carcinoma. ZNF608 and CSF1R may be used as predictors of radiosensitivity of nasopharyngeal carcinoma. (4) there is no correlation between the expression of ZNF608 and CSF1R in nasopharyngeal carcinoma tissue and age, clinical stage, T stage and cervical lymph node metastasis.
