【摘要】：目的檢測CHD5與上皮鈣粘蛋白(E-cad)在鱗狀細胞喉癌(LSCC)組織中的表達,探討其與分期、淋巴結(jié)轉(zhuǎn)移及預(yù)后的關(guān)系,并分析兩者的相關(guān)性。方法通過免疫組織化學(xué)法和Western印跡法檢測103例喉癌患者中,CHD5和E-cadherin蛋白的表達水平,并分析二者對喉癌患者預(yù)后的影響。結(jié)果 CHD5和E-cad蛋白在喉癌組織表達水平顯著低于癌旁正常組織(P0.05),二者的表達水平與LSCC患者的TNM分期、淋巴結(jié)轉(zhuǎn)移及預(yù)后呈顯著負相關(guān),而與患者的性別、年齡因素?zé)o相關(guān)性。CHD5蛋白與E-cad蛋白的表達的平均光密度值(AOD)呈顯著正相關(guān)(r=0.814,P0.01)。結(jié)論 CHD5基因可能通過調(diào)控E-cad蛋白的表達參與了喉癌上皮間質(zhì)轉(zhuǎn)化的過程。
[Abstract]:Objective to detect the expression of CHD5 and E-cadin in squamous cell laryngeal carcinoma (SCC), to investigate the relationship between the expression of E-cadand and staging, lymph node metastasis and prognosis, and to analyze the correlation between them. Methods Immunohistochemistry and Western blotting were used to detect the expression of CHD5 and E-cadherin protein in 103 patients with laryngeal carcinoma. Results the expression of CHD5 and E-cad protein in laryngeal carcinoma was significantly lower than that in the adjacent normal tissue (P 0.05). The expression level of CHD5 and E-cad was negatively correlated with the TNM stage, lymph node metastasis and prognosis of LSCC patients, but negatively correlated with the sex of the patients. There was no correlation between age factors and the average optical density of E-cad protein expression. There was a significant positive correlation between CHD5 protein and E-cad protein expression. Conclusion CHD5 gene may be involved in the process of epithelial interstitial transformation of laryngeal carcinoma by regulating the expression of E-cad protein.
