糖尿病大鼠視網(wǎng)膜組織醛脫氫酶活性與表達量變化及其意義

【摘要】:目的探討醛脫氫酶(ALDH)在糖尿病視網(wǎng)膜病變發(fā)生、發(fā)展中的作用。方法選擇SD大鼠40只,隨機分為模型組、對照組各20只。模型組采用腹腔注射鏈脲佐菌素建立糖尿病模型,對照組同期腹腔注射等體積檸檬酸鈉緩沖液。兩組分別于建模3天及建模8、16周檢測體質(zhì)量及血糖;建模8、16周各處死10只,檢測視網(wǎng)膜組織ALDH活性和表達量,采用免疫組化法檢測視網(wǎng)膜組織bcl-2、bax表達,采用TUNEL法檢測視網(wǎng)膜神經(jīng)細胞凋亡指數(shù)。結(jié)果兩組體質(zhì)量隨建模時間延長均明顯增加(P均0.05),但血糖變化不明顯;與對照組比較,模型組建模8、16周體質(zhì)量明顯低于對照組同期(P均0.05),而血糖水平明顯高于對照組同期(P均0.05)。與對照組比較,模型組建模8、16周ALDH活性和表達量均明顯升高(P均0.05);模型組建模16周ALDH活性和表達量均低于建模8周(P均0.05);對照組建模8、16周ALDH活性和表達量變化不明顯(P均0.05)。模型組建模8、16周bcl-2、bax表達及視網(wǎng)膜神經(jīng)細胞凋亡率均高于對照組同期,且建模16周明顯高于建模8周(P均0.05),而對照組變化不明顯(P0.05)。結(jié)論糖尿病大鼠視網(wǎng)膜組織ALDH活性與表達量升高,其活性與表達量變化可促進糖尿病大鼠視網(wǎng)膜神經(jīng)細胞凋亡,且與凋亡嚴重程度有關(guān)。
[Abstract]:Objective to investigate the role of aldehydes dehydrogenase (ALDH) in the pathogenesis and development of diabetic retinopathy. Methods Forty SD rats were randomly divided into two groups: the model group (n = 20) and the control group (n = 20). The diabetic model was established by intraperitoneal injection of streptozotocin, while the control group was intraperitoneally injected with sodium citrate buffer. The body mass and blood glucose were detected in the two groups after 3 days and 816 weeks respectively, and 10 rats were killed in each group at 816 weeks. The activity and expression of ALDH in retinal tissues were detected, and the expression of bcl-2mbax in retinal tissues was detected by immunohistochemical method, and the expression of bcl-2mbax in retina was detected by immunohistochemical method. Apoptosis index of retinal nerve cells was measured by TUNEL method. Results compared with the control group, the body mass of the model group at 816 weeks was significantly lower than that of the control group, and the blood glucose level was significantly higher than that of the control group. Compared with the control group, the activity and expression of ALDH in the model group increased significantly at 816 weeks, the activity and expression of ALDH in the model group at the 16th week were lower than those in the model group at the 8th week (P 0.05), and the ALDH activity and expression in the control group were not significantly changed at 816 weeks after modeling (P < 0.05). The expression of bcl-2mbax and the rate of apoptosis of retinal nerve cells in the model group were significantly higher than those in the control group at 8 and 16 weeks, and were significantly higher than those in the model group at the 8th week and the control group respectively, but the changes in the control group were not obvious. Conclusion the activity and expression of ALDH in the retina of diabetic rats were increased, and the changes of the activity and expression of ALDH could promote the apoptosis of retinal nerve cells in diabetic rats, which was related to the severity of apoptosis.
