【摘要】：目的探討miR-34b在老年鼻咽癌組織中表達(dá)及對細(xì)胞增殖、侵襲的影響。方法選取初治原發(fā)性老年鼻咽癌患者83例,正常鼻咽黏膜組織40例作為對照組,培養(yǎng)人鼻咽癌CNE-2細(xì)胞株,根據(jù)轉(zhuǎn)染物不同將細(xì)胞分為miR-34b轉(zhuǎn)染組、陰性對照組和空白對照組,利用實時熒光定量PCR技術(shù)檢測老年鼻咽癌及對照組組織中miR-34b表達(dá)以及不同轉(zhuǎn)染組細(xì)胞中miR-34b表達(dá),MTT比色法檢測不同轉(zhuǎn)染組細(xì)胞增殖能力,檢測細(xì)胞黏附能力,Transwell法檢測不同轉(zhuǎn)染組細(xì)胞侵襲能力。結(jié)果老年鼻咽癌組織中miR-34b相對表達(dá)量為(0.64±0.09),顯著低于對照組的(0.94±0.12),差異有統(tǒng)計學(xué)意義(t=14.410,P0.001);miR-34b在老年鼻咽癌組織中相對表達(dá)量與臨床分期、T分期、N分期和淋巴結(jié)轉(zhuǎn)移有關(guān)(P0.001);miR-34b轉(zhuǎn)染組細(xì)胞中miR-34b相對表達(dá)量為(0.85±0.10),顯著高于陰性對照組和空白對照組,分別為(0.52±0.08)和(0.50±0.07),差異有統(tǒng)計學(xué)意義(F=243.329,P0.001);與陰性對照組和空白對照組相比,miR-34b轉(zhuǎn)染組細(xì)胞增殖能力顯著降低(P0.05);miR-34b轉(zhuǎn)染組黏附細(xì)胞數(shù)顯著高于陰性對照組和空白對照組,而侵襲細(xì)胞數(shù)顯著低于陰性對照組和空白對照組(P0.001)。結(jié)論 miR-34b在老年鼻咽癌組織中呈低表達(dá),上調(diào)miR-34b表達(dá)可減少細(xì)胞增殖,增加細(xì)胞黏附能力,抑制細(xì)胞侵襲能力,有望成為提高鼻咽癌對放射治療敏感性的目的基因。
[Abstract]:Objective to investigate the expression of miR-34b in the tissues of elderly nasopharyngeal carcinoma (NPC) and the effect on cell proliferation and invasion. Methods 83 patients with primary nasopharyngeal carcinoma and 40 normal nasopharyngeal mucosa tissues were selected as control group, and the human nasopharyngeal carcinoma CNE-2 cell lines were cultured. The cells were divided into miR-34b transfection group, negative control group and blank control group according to the different transfection. The expression of miR-34b in the elderly nasopharyngeal carcinoma and the control group and the expression of miR-34b in the different transfected groups were detected by real time fluorescence quantitative PCR. The cell proliferation ability of different transfected groups was detected by MTT colorimetric assay and the cell adhesion ability was detected by the Transwell method. The results of m in the old nasopharyngeal carcinoma tissues were determined by Transwell. The relative expression of iR-34b was (0.64 + 0.09), significantly lower than that of the control group (0.94 + 0.12), and the difference was statistically significant (t=14.410, P0.001). The relative expression of miR-34b in the old nasopharyngeal carcinoma tissues was related to the clinical stage, T stage, N staging and lymph node metastasis (P0.001), and the relative expression of miR-34b in the cells of miR-34b transfected group was (0.85 + 0.10). Compared with negative control group and blank control group (0.52 + 0.08) and (0.50 + 0.07), the difference was statistically significant (F=243.329, P0.001). Compared with the negative control group and the blank control group, the cell proliferation ability of miR-34b transfected group was significantly decreased (P0.05), and the number of adhesion cells in miR-34b transfer group was significantly higher than that of the negative control group and the blank control group, and the number of cells in the miR-34b transfer group was significantly higher than that in the negative control group and the blank control group. The number of invasive cells was significantly lower than that of the negative control group and the blank control group (P0.001). Conclusion miR-34b is low expression in the elderly nasopharyngeal carcinoma tissue. Up regulation of miR-34b expression can reduce cell proliferation, increase cell adhesion ability and inhibit cell invasiveness. It is expected to be the target gene to improve the sensitivity of nasopharyngeal carcinoma to radiotherapy.
