原花青素對氧化應激人眼小梁網(wǎng)細胞凋亡及細胞色素C釋放的影響

【摘要】:目的研究原花青素對H2O2誘導人眼小梁網(wǎng)細胞(human trabecular meshwork cells,HTMC)凋亡及細胞色素C釋放的影響。方法將HTMC進行傳代后隨機分為5組。未處理組:正常培養(yǎng)的HTMC;對照組:正常培養(yǎng)的HTMC+H_2O_2(500μmol·L~(-1)處理1 h);3個濃度的原花青素組:正常培養(yǎng)的HTMC+H_2O_2(500μmol·L~(-1)處理1 h)+原花青素(濃度分別為0.02 g·L~(-1)、0.05 g·L~(-1)、0.10 g·L~(-1))。使用Annexin V-FITC檢測細胞凋亡情況,Western blot檢測HTMC胞漿細胞色素C含量。結(jié)果與未處理組相比,對照組及各原花青素組細胞凋亡率均升高,差異均有統(tǒng)計學意義(均為P0.01)。與對照組相比,各原花青素組細胞凋亡率均降低,差異均有統(tǒng)計學意義(均為P0.01);不同濃度原花青素組間隨原花青素濃度增加細胞凋亡率下降,各原花青素組間差異均有統(tǒng)計學意義(均為P0.01)。與未處理組相比,對照組以及0.02 g·L~(-1)和0.05 g·L~(-1)原花青素組細胞色素C的釋放均增加,差異均有統(tǒng)計學意義(均為P0.01);而0.10 g·L~(-1)原花青素組與未處理組相比,差異無統(tǒng)計學意義(P0.05)。與對照組相比,各原花青素組細胞色素C釋放均減少,差異均有統(tǒng)計學意義(均為P0.01)。不同濃度原花青素組間隨原花青素濃度增加細胞色素C的釋放減少,各組間差異均有統(tǒng)計學意義(均為P0.01)。結(jié)論外源性原花青素可以降低氧化應激HTMC的凋亡率,減少細胞色素C的釋放,有較強的抗氧化作用,并在一定濃度范圍內(nèi)隨著濃度的增加其抗氧化作用逐漸增強。
[Abstract]:Objective to study the effect of procyanidins on apoptosis and cytochrome C release in human trabecular meshwork cells (HTMC) induced by H2O2. Methods HTMC were randomly divided into 5 groups after passage. Untreated group: normal culture HTMCs; control group: normal cultured HTMC H_2O_2(500 渭 mol L ~ (1) treated for 1 hour; 3 concentration of proanthocyanidins group: normal culture HTMC H_2O_2(500 渭 mol L ~ (-1) for 1 h) procyanidins (0.02g / L ~ (-1) (0.02g / L ~ (-1) ~ (-0.10) g / L ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1). Apoptosis was detected by Annexin V-FITC and cytochrome C in cytoplasm of HTMC was detected by Western blot. Results compared with the untreated group, the apoptotic rate of the control group and the proanthocyanidins group were significantly higher than that of the untreated group (P 0.01). Compared with the control group, the apoptotic rate of each proanthocyanidin group was significantly lower than that of the control group (P 0.01), and the apoptosis rate decreased with the increase of proanthocyanidin concentration among different concentrations of proanthocyanidins. The difference of proanthocyanidin group was statistically significant (P 0.01). Compared with untreated group, the release of cytochrome C in the control group and 0.02 g / L ~ (-1) and 0.05 g / L ~ (-1) proanthocyanidins group was significantly higher than that in the untreated group (all P 0.01 ~ (-1); 0.10 g / L ~ (-1) there was no significant difference between the procyanidin group and the untreated group (P 0.05). Compared with the control group, the release of cytochrome C in each procyanidin group was significantly lower than that in the control group (P 0.01). With the increase of proanthocyanidin concentration, the release of cytochrome C decreased in different concentrations of proanthocyanidins, and the difference was statistically significant (P 0.01). Conclusion exogenous proanthocyanidins can reduce the apoptosis rate of oxidative stress HTMC and reduce the release of cytochrome C.
