金屬硫蛋白與大鼠放射性白內(nèi)障的關(guān)系

【摘要】:目的: 利用動(dòng)物實(shí)驗(yàn),通過一次性大劑量(25Gy) X射線照射S-D大鼠雙眼建立放射性白內(nèi)障模型,觀察照射后一周、二周、四周時(shí)大鼠晶狀體情況,同時(shí)檢測(cè)其晶狀體金屬硫蛋白(Metallothionein,MT)的表達(dá)水平,探討金屬硫蛋白與放射性白內(nèi)障形成之間的關(guān)系。 方法: 照射前選取8只大鼠作為正常對(duì)照組。實(shí)驗(yàn)照射組大鼠24只,一次性照射X射線(雙眼同時(shí)照射)25Gy。所有大鼠分別于照射后一周、二周、四周時(shí),腹腔注射3.6%水合氯醛麻醉后,用托吡卡胺滴眼液充分散瞳,裂隙燈顯微鏡觀察并記錄晶狀體混濁情況;然后在上述每個(gè)時(shí)間點(diǎn)的實(shí)驗(yàn)照射組依次隨機(jī)選取8只大鼠處死取其晶狀體。分別運(yùn)用逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(Reverse Transcription-Polymerase Chain Reaction,RT-PCR)和免疫印跡法(Western blotting)檢測(cè)MT-Ⅰ、MT-ⅡmRNA和金屬硫蛋白(MT)在上述時(shí)間點(diǎn)大鼠晶狀體內(nèi)的表達(dá)情況。 應(yīng)用SPSS17.0統(tǒng)計(jì)軟件對(duì)結(jié)果進(jìn)行統(tǒng)計(jì)分析,采用Independent Samples T-Test法和One-Way-ANOVA法分別對(duì)計(jì)量資料進(jìn)行獨(dú)立樣本T檢驗(yàn)和單因素方差分析,并使用LSD法進(jìn)行組間比較。 結(jié)果: 1.形態(tài)學(xué):一次性大劑量(25Gy)X射線照射大鼠雙眼,于照射后一周、二周時(shí)散瞳觀察大鼠晶狀體均未見混濁,而照射后四周觀察時(shí)可見晶狀體后囊和后皮質(zhì)開始出現(xiàn)細(xì)小、散在白點(diǎn)狀混濁。 2.RT-PCR結(jié)果顯示:在mRNA水平MT-Ⅰ、MT-Ⅱ在大鼠晶狀體上均有表達(dá),實(shí)驗(yàn)組和對(duì)照組的晶狀體進(jìn)行RT-PCR均可獲得MT-Ⅰ、MT-Ⅱ的陽(yáng)性目的基因。實(shí)驗(yàn)組晶狀體的MT-Ⅰ、MT-ⅡmRNA的表達(dá)均較對(duì)照組均有不同程度的增加,差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。其變化是隨著照射后時(shí)間的延長(zhǎng),上述時(shí)間點(diǎn)MT-Ⅰ、MT-ⅡmRNA的表達(dá)呈先上升后下降的趨勢(shì),各時(shí)間點(diǎn)的變化有差異,差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。 3. Western blotting結(jié)果顯示:實(shí)驗(yàn)照射組金屬硫蛋白含量相對(duì)于實(shí)驗(yàn)對(duì)照組增加,差異有統(tǒng)計(jì)學(xué)意義(P<0.05);實(shí)驗(yàn)照射組金屬硫蛋白含量的變化隨照射時(shí)間的延長(zhǎng)呈先增加后降低的趨勢(shì),并且上述時(shí)間點(diǎn)的變化有差異,其差異有顯著統(tǒng)計(jì)學(xué)意義(P<0.05)。 結(jié)論: 一次性大劑量X射線(25Gy)照射大鼠雙眼可在短時(shí)間內(nèi)即可引起晶體混濁;同時(shí)它可提高晶狀體MT-Ⅰ、MT-ⅡmRNA及MT蛋白的表達(dá)水平;當(dāng)MT表達(dá)下降時(shí)晶體出現(xiàn)混濁,說明MT可能參與放射性白內(nèi)障的形成。
[Abstract]:Objective: In animal experiment, the model of radiation-induced cataract was established in the eyes of S-D rats by X-ray irradiation with a large dose of 25 Gy at one time. The lens of the rats was observed one week, two weeks and four weeks after irradiation. At the same time, the expression level of metallothionein MTA was detected to explore the relationship between metallothionein and radiation cataract formation. Methods: Eight rats were selected as normal control group before irradiation. Twenty-four rats in the experimental group were exposed to X ray at one time (25 Gy for both eyes at the same time). All the rats were injected intraperitoneally with 3.6% chloral hydrate anesthetized one week, two weeks and four weeks after irradiation. The lens opacity was observed and recorded by slit lamp microscope. Then 8 rats were randomly selected at each time point in the experimental group to get their lens.
