靶向DC DNA疫苗的研制及抗鼠眼角膜HSV-1感染的初步研究

【摘要】:目的:構(gòu)建靶向樹突狀細(xì)胞(DC)的DNA疫苗pRSC-NLDC145·gD-IL-21,針對抵抗鼠眼角膜單純皰疹病毒1型(HSV-1)的感染,探討其是否能產(chǎn)生更強(qiáng)的免疫應(yīng)答反應(yīng)。方法:通過基因克隆的方法構(gòu)建靶向DC的重組質(zhì)粒pRSC-NLDC145·gD-IL-21,對其進(jìn)行鑒定,并和殼聚糖包裹組成納米DNA疫苗,分別以靶向組pRSC-NLDC145·gD-IL-21+殼聚糖、非靶向組pRSC-gD-IL-21+殼聚糖、pRSC+殼聚糖和殼聚糖免疫小鼠3次,間隔2周,檢測各疫苗對小鼠免疫應(yīng)答的水平。結(jié)果:靶向DC的重組質(zhì)?稍谡婧思(xì)胞內(nèi)進(jìn)行蛋白水平的表達(dá),且和納米殼聚糖包裹形成包封率較高的靶向DNA疫苗組,較非靶向組在鼠體內(nèi)產(chǎn)生了更強(qiáng)的免疫應(yīng)答反應(yīng)。結(jié)論:眼表黏膜接種靶向DC的納米DNA疫苗pRSC-NLDC145·gD-IL-21,在小鼠單純皰疹病毒性角膜炎(HSK)動物模型中顯示了更強(qiáng)的免疫保護(hù)反應(yīng)。
[Abstract]:Aim: to construct a DNA vaccine pRSC-NLDC145 gD-IL-21 targeting dendritic cells (DC) and investigate whether it can produce a stronger immune response to the infection of corneal herpes simplex virus type 1 (HSV-1) in mice. Methods: the recombinant plasmid pRSC-NLDC145 gD-IL-21 was constructed by gene cloning. The recombinant plasmid pRSC-NLDC145 gD-IL-21 was identified and encapsulated with chitosan to form a DNA vaccine. PRSC-NLDC145 gD-IL-21 chitosan was used as the target group. Mice were immunized with pRSC-gD-IL-21 chitosan and chitosan for 3 times, at intervals of 2 weeks, to detect the level of immune response of each vaccine to mice. Results: the recombinant plasmid targeting DC could express protein level in eukaryotic cells, and encapsulated with nano-chitosan to form a higher encapsulation efficiency of targeted DNA vaccine group, which had a stronger immune response than non-target group in mice. Conclusion: pRSC-NLDC145 gD-IL-21, a nano-sized DNA vaccine targeting DC, showed a stronger immune protective response in mice with herpes simplex keratitis.
