微小RNA-140-5p在老年下咽癌組織中的表達(dá)及對下咽癌細(xì)胞株FaDu遷移、侵襲力的影響

【摘要】:目的探討微小RNA-140-5p(MiR-140-5p)影響去整合素金屬蛋白酶(ADAM)10表達(dá)對老年下咽癌組織和細(xì)胞的作用。方法實(shí)時熒光定量RCR檢測MiR-140-5p和ADAM10在41例老年下咽癌患者腫瘤組織及癌旁組織中的表達(dá);利用轉(zhuǎn)染技術(shù)分別上調(diào)MiR-140-5p表達(dá),下調(diào)MiR-140-5p和ADAM10表達(dá);采用Transwell細(xì)胞侵襲/遷移實(shí)驗(yàn)檢測MiR-140-5p上調(diào)、MiR-140-5p下調(diào)、ADAM10下調(diào)對FaDu細(xì)胞遷移、侵襲的影響;Western印跡檢測41對組織標(biāo)本和FaDu細(xì)胞的ADAM10蛋白表達(dá)。結(jié)果 MiR-140-5p在下咽癌組織中明顯下調(diào),ADAM10 mRNA和蛋白在下咽癌組織中均明顯上調(diào)。實(shí)驗(yàn)組FaDu細(xì)胞干擾ADAM10表達(dá)后,ADAM10表達(dá)水平明顯低于陰性對照組(P0.05)。Transwell實(shí)驗(yàn)顯示,遷移和侵襲實(shí)驗(yàn)中MiR-140-5p上調(diào)組每視野下平均遷移細(xì)胞數(shù)均明顯低于對照組,MiR-140-5p下調(diào)組均明顯高于對照組(P0.05);ADAM10干擾實(shí)驗(yàn)組遷移和侵襲細(xì)胞數(shù)明顯低于對照組(P0.05)。ADAM10下調(diào)可逆轉(zhuǎn)MiR-140-5p下調(diào)對細(xì)胞遷移和侵襲能力的促進(jìn)作用。Western印跡顯示,MiR-140-5p表達(dá)上調(diào)后,實(shí)驗(yàn)組ADAM10蛋白表達(dá)水平明顯低于陰性對照組;MiR-140-5p表達(dá)下調(diào)后,實(shí)驗(yàn)組ADAM10蛋白表達(dá)水平明顯高于陰性對照組。結(jié)論 MiR-140-5p在老年下咽癌組織中低表達(dá),ADAM10高表達(dá)。MiR-140-5p抑制下咽癌細(xì)胞遷移、侵襲力可通過下調(diào)ADAM10蛋白表達(dá)實(shí)現(xiàn),MiR-140-5p可作為下咽癌治療的新靶點(diǎn)。
[Abstract]:Objective to investigate the effect of microRNA-140-5pmMiR-140-5p) on the expression of desintegrin metalloproteinase (ADAMn10) in elderly patients with hypopharyngeal carcinoma. Methods Real-time quantitative RCR was used to detect the expression of MiR-140-5p and ADAM10 in 41 elderly patients with hypopharyngeal carcinoma, and the expression of MiR-140-5p was up-regulated and the expression of MiR-140-5p and ADAM10 was down-regulated by transfection technique. Transwell cell invasion / migration assay was used to detect the up-regulation of MiR-140-5p by MiR-140-5p down-regulating the down-regulation of ADAM10 on the migration of FaDu cells. Western blot was used to detect the expression of ADAM10 protein in 41 tissues and FaDu cells. Results MiR-140-5p significantly down-regulated ADAM10 mRNA and up-regulated protein in hypopharyngeal carcinoma tissues. The level of ADAM10 expression in FaDu cells of experimental group was significantly lower than that in negative control group after interfering with ADAM10 expression. In the migration and invasion experiment, the average number of migration cells in the up-regulated group of MiR-140-5p was significantly lower than that in the down-regulated group of MiR-140-5p in the control group, which was significantly higher than that in the control group in which P0.05ADAM10 interfered with migration and invasion cells. The down-regulation of P0.05U. ADAM10 in the experimental group was significantly lower than that in the control group. Effects of MiR-140-5p down-regulation on cell migration and invasion. Western blot showed that MiR-140-5p expression was up-regulated. The expression of ADAM10 protein in the experimental group was significantly lower than that in the negative control group after down-regulation of MiR-140-5p, and the expression level of ADAM10 protein in the experimental group was significantly higher than that in the negative control group. Conclusion the low expression of ADAM10 and the high expression of MiR-140-5p in elderly hypopharyngeal carcinoma can inhibit the migration of hypopharyngeal carcinoma cells. The invasiveness can be realized by down-regulating the expression of ADAM10 protein. MiR-140-5p can be used as a new target for the treatment of hypopharyngeal carcinoma.
