喉癌中鈣網(wǎng)織蛋白和CD47的表達及其與T淋巴細胞浸潤的相關(guān)性

【摘要】:目的:通過檢測喉鱗狀上皮細胞癌(laryngeal squamous cell carcinoma,LSCC)組織中促吞噬信號作用的鈣網(wǎng)織蛋白(calreticulin,CRT)和抗吞噬信號作用CD47的表達,探討二者在LSCC發(fā)生發(fā)展中的作用及其與T淋巴細胞浸潤的相關(guān)性。方法:應用逆轉(zhuǎn)錄酶-聚合酶鏈反應(RT-PCR)檢測42例LSCC組織及18例癌旁正常鱗狀上皮組織中CRT和CD47mRNA的表達水平,免疫組化S-P法檢測68例原發(fā)性LSCC及20例癌旁正常組織中CRT和CD47蛋白的表達情況,并用CD3及CD45RO單克隆抗體標記癌組織實質(zhì)及間質(zhì)中T淋巴細胞浸潤情況。結(jié)果:在LSCC組織中,CRT和CD47mRNA和蛋白的表達均高于癌旁正常鱗狀上皮組織(P0.05);CRT及CD47蛋白隨著LSCC的腫瘤T分期、淋巴結(jié)轉(zhuǎn)移及臨床分期的增加其表達增加(P0.05),CD47與CRT蛋白的表達有一種較強的正相關(guān)性(r=0.89P0.05);CRT蛋白的表達與CD45RO+T淋巴細胞的高浸潤相關(guān)(P0.05),CD47蛋白的表達與CD3+T淋巴細胞及癌組織間質(zhì)中CD45RO+T淋巴細胞的浸潤無相關(guān)性(P0.05). 結(jié)論:CRT和CD47在LSCC的發(fā)生、發(fā)展過程中可起重要作用,,CRT的表達與CD45RO+T淋巴細胞的高浸潤相關(guān),可能是喉鱗癌的一種新的抗腫瘤免疫機制。
[Abstract]:Objective: to detect the expression of calreticulinus calreticulinus (CRT) and CD47 in laryngeal squamous cell carcinoma of larynx squamous cell carcinoma (LSCC).To explore the role of both of them in the pathogenesis and development of LSCC and the correlation between them and T lymphocyte infiltration.Methods: reverse transcriptase polymerase chain reaction (RT-PCR) was used to detect the expression of CRT and CD47mRNA in 42 cases of LSCC and 18 cases of adjacent normal squamous epithelium.Immunohistochemical S-P method was used to detect the expression of CRT and CD47 protein in 68 cases of primary LSCC and 20 cases of adjacent normal tissues. T-lymphocyte infiltration in parenchyma and interstitial tissue was labeled with CD3 and CD45RO monoclonal antibodies.Results: the expression of CRT, CD47mRNA and protein in LSCC was higher than that in normal squamous epithelium adjacent to the carcinoma (P0.05) and CD47 protein was associated with the T stage of LSCC.The expression of CD47 and CRT protein in lymph node metastasis and clinical stage increased. There was a strong positive correlation between the expression of CD47 and CRT protein and the high infiltration of CD45RO T lymphocytes. The expression of CD47 protein was correlated with the expression of CD3 T lymphocytes and carcinoma.There was no correlation between the infiltration of CD45RO T lymphocytes in the interstitial tissue (P 0.05).Conclusion the expression of LSCC and CD47 may play an important role in the pathogenesis and development of LSCC. The expression of CRT may be associated with the high infiltration of CD45RO T lymphocytes, which may be a new anti-tumor immune mechanism of laryngeal squamous cell carcinoma.
