銀杏葉提取物對(duì)慢性間歇性低氧大鼠胸主動(dòng)脈重構(gòu)的影響

【摘要】:目的:探索銀杏葉提取物對(duì)慢性間歇性低氧大鼠胸主動(dòng)脈血管重構(gòu)的作用。方法:選取30只健康、雄性Wistar大鼠,隨機(jī)分為正常對(duì)照組(NC組)、慢性間歇性低氧組(CIH組)、慢性間歇低氧聯(lián)合銀杏葉提取物灌胃組(CIH+GBE組),各組10只。HE染色法檢測(cè)各試驗(yàn)大鼠胸主動(dòng)脈血管的病理變化;用熒光定量PCR(qRTPCR)方法檢測(cè)胸主動(dòng)脈p38MAPK、ERK1/2、JNK的mRNA表達(dá)水平;用Western blot方法檢測(cè)上述三個(gè)因子蛋白表達(dá)水平及p-ERK1/2的水平;用免疫酶聯(lián)反應(yīng)法檢測(cè)大鼠血清中腫瘤壞死因子α(TNF-α)、白細(xì)胞介素6(IL-6)、超敏C反應(yīng)蛋白(hsCRP)的含量。結(jié)果:(1)HE染色病理結(jié)果顯示:與NC組相比CIH組胸主動(dòng)脈出現(xiàn)重構(gòu),胸主動(dòng)脈見中膜厚度增加、中膜厚度與管腔內(nèi)徑之比增大(P0.05),CIH+GBE組與CIH組相比,上述兩指標(biāo)均降低(P0.05),Masson染色顯示:與NC組相比CIH組胸主動(dòng)脈膠原纖維化面積明顯增加(P0.05),而CIH+GBE組與CIH組相比,上述指標(biāo)均降低(P0.05);(2)qRT-PCR檢測(cè)結(jié)果顯示:與NC組比較IH組胸主動(dòng)脈中p38MAPK、JNK的mRNA轉(zhuǎn)錄水平明顯增強(qiáng)(P0.05),CIH+GBE與IH組比較發(fā)現(xiàn)CIH+GBE組p38MAPK、JNK的mRNA變化趨勢(shì)與上一組比較相反(P0.05),而ERK1/2的轉(zhuǎn)錄水平在上述兩組中比較無明顯變化(P0.05);(3)Westen blot檢查顯示:各組p38MAPK、JNK、總ERK1/2蛋白變化趨勢(shì)與qRTPCR檢查結(jié)果一致,而與NC組相比CIH組ERK1/2磷酸化明顯升高(P0.05),CIH+GBE組與CIH組比較,CIH+GBE組ERK1/2磷酸化水平明顯降低(P0.05);(4)ELISA檢測(cè)結(jié)果顯示:與NC組相比,CIH組大鼠血清中TNF-α、IL-6、hsCRP因子的水平均顯著上升(P0.05),而CIH+GBE組與CIH組比較,大鼠血清中TNF-α、hsCRP的含量明顯降低(P0.05),IL-6有所降低,但無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:間歇性低氧可以引起大鼠胸主動(dòng)脈血管重構(gòu);銀杏葉提取物可以抑制間歇性缺氧大鼠胸主動(dòng)脈血管ERK1/2蛋白磷酸化,降低大鼠主動(dòng)脈表達(dá)JNK、p38MAPK,并降低其血清中TNF-α、hsCRP的水平。
[Abstract]:Objective: to investigate the effect of ginkgo biloba extract on vascular remodeling of thoracic aorta in chronic intermittent hypoxia rats.Methods: 30 healthy male Wistar rats were selected.They were randomly divided into normal control group (n = 10), chronic intermittent hypoxia group (n = 10), chronic intermittent hypoxia group (n = 10), chronic intermittent hypoxia group (n = 10), chronic intermittent hypoxia group (n = 10) and ginkgo biloba extract (Ginkgo biloba extract) group (n = 10).The expression of mRNA in the thoracic aorta p38 MAPKG ERK1 / 2 JNK was detected by fluorescence quantitative PCRQ RT PCR, and the expression levels of the above three factors and p-ERK1/2 were detected by Western blot method.The levels of tumor necrosis factor- 偽 (TNF- 偽), interleukin-6 (IL-6) and hypersensitive C-reactive protein (hsCRP) in the serum of rats were detected by immuno-enzyme linked reaction (Elisa).Results compared with NC group, there were remodeling of thoracic aorta, increase of medial thickness of thoracic aorta and increase of ratio of medial thickness to lumen diameter in CIH group compared with CIH group.Compared with NC group, the area of collagen fibrosis of thoracic aorta in CIH group was significantly increased, while that in CIH GBE group was significantly higher than that in CIH group, and that in CIH GBE group was significantly higher than that in CIH group.The results of RT-PCR showed that compared with NC group, the mRNA transcription level of p38MAPK-1 JNK in thoracic aorta of IH group was significantly enhanced than that of IH group and IH group. It was found that the mRNA change trend of p38MAPKN JNK in CIH GBE group was opposite to that in the previous group (P0.05), but ERK1/2 translocation in IH group was different from that in the previous group.There was no significant change in the recorded level in the above two groups. The results of blot showed that the change trend of total ERK1/2 protein in each group was consistent with that of qRTPCR.CIH GBE group was compared with CIH group.The content of TNF- 偽 hsCRP in the serum of rats was significantly decreased, but the level of IL-6 was decreased, but there was no significant difference in serum TNF- 偽 hsCRP.Conclusion: intermittent hypoxia can induce vascular remodeling of thoracic aorta in rats, Ginkgo biloba extract can inhibit the phosphorylation of ERK1/2 protein in thoracic aorta of rats with intermittent hypoxia, decrease the expression of ERK1/2 p38 MAPK in rat aorta, and decrease the level of TNF- 偽 hsCRP in serum.
