【摘要】：目的:探討白細(xì)胞介素19(interleukin-19,IL-19)及其受體(IL-20R1/IL-20R2)在不同類型慢性鼻-鼻竇炎[慢性鼻-鼻竇炎伴鼻息肉(chronic rhinosinusitis with nasal polyps,CRSw NP)和慢性鼻-鼻竇炎不伴鼻息肉(chronic rhinosinusitis without nasal polyps,CRSs NP)]患者中表達(dá)的差異,并分析其與慢性鼻-鼻竇炎組織重塑的相關(guān)性。方法:研究共分3組:CRSw NP組(30例)、CRSs NP組(15例)和對照組(鼻中隔偏曲患者15例)。實(shí)時熒光定量PCR檢測IL-19及其受體(IL-20R1/IL-20R2)和組織重塑因子基質(zhì)金屬蛋白酶(MMP)-2、MMP-9、金屬蛋白酶組織抑制物(TIMP)-1的mRNA相對表達(dá)量;免疫組化檢測IL-19及其受體的蛋白表達(dá)量;ELISA檢測MMP-9、TIMP-1的蛋白表達(dá)量。結(jié)果:CRSw NP組IL-19、IL-20R1、IL-20R2和MMP-9的mRNA和蛋白表達(dá)量高于CRSs NP組和對照組(P0.05),CRSs NP組TIMP-1的mRNA和蛋白的表達(dá)量均高于CRSw NP組和對照組(P0.05);MMP-2的mRNA和蛋白的表達(dá)量在各組無顯著差異。CRSw NP組IL-19、IL-20R1和IL-20R2均與MMP-9的mRNA相對表達(dá)量呈正相關(guān)(P0.05);IL-19及其受體與TIMP-1的mRNA相對表達(dá)量無明顯相關(guān)。結(jié)論:IL-19及其受體的mRNA在CRSw NP中高表達(dá),且與MMP-9 mRNA表達(dá)呈正相關(guān),提示二者可能存在交互作用,可能參與慢性鼻-鼻竇炎的組織重塑。
[Abstract]:Objective: to investigate the expression of interleukin-19 interleukin-19 (IL-19) and its receptor IL-20R1 / IL-20R2 in patients with chronic rhinosinusitis [chronic rhinosinusitis with chronic rhinosinusitis with nasal polyps] and chronic rhinosinusitis without chronic rhinosinusitis without nasal polyps (CRSNPs). Methods: the study was carried out in 30 cases of 30 cases of CRSs NP group and 15 cases of control group (15 cases of nasal septum deviation patients). Real-time fluorescence quantitative PCR was used to detect IL-19 and its receptor in patients with chronic rhinosinusitis and chronic rhinosinusitis. MRNA expression of IL-20R1 / IL-20R2), MMP-2MMP-2MMP-9, tissue inhibitor of metalloproteinase (TIMP- 1), tissue remodeling factor (MMP-2MMP-2MMP 9), tissue inhibitor of metalloproteinase (TIMP- 1); The protein expression of IL-19 and its receptor was detected by immunohistochemistry and the protein expression of MMP-9 and TIMP-1 was detected by Elisa. Results the mRNA and protein expression of IL-19, IL-20R1, IL-20R2 and MMP-9 in the group of IL-19 / CRSw NP was higher than that in the group of CRSs NP and control group (P 0.05) and the expression of TIMP-1 protein was higher than that in the group of CRSw. There was no significant difference in the expression of mRNA and protein between NP group and control group. There was no significant correlation between the expression of IL-19 IL-20R1 and IL-20R2 and the relative expression of mRNA of MMP-9 in CRSw NP group. Conclusion there is no significant correlation between the expression of IL-19 and its receptor and the relative expression of mRNA of TIMP-1 in the NP group and the control group. Conclusion\\\; Somatic mRNA was overexpressed in CRSw NP. There was a positive correlation with the expression of MMP-9 mRNA, which suggested that there might be interaction between the two proteins, which might be involved in the tissue remodeling of chronic rhinosinusitis.
