靈長類動物自發(fā)性年齡相關眼病動物模型的探討

【摘要】:目的 本研究篩查老年猴群中自然產生的白內障,年齡相關性黃斑變性,糖尿病視網膜病變等眼病,通過對比靈長類動物眼底疾病改變與人類眼底疾病改變,探討在老年猴群中建立年齡相關性眼病實驗動物模型的可行性。方法 39只食蟹猴,平均年齡(11.9±2.05)歲,最小9歲,最大17歲和13只獼猴,平均年齡(15.5±5.2)歲,最小7歲,最大24歲,進行空腹血糖和2 h糖耐量檢測。應用裂隙燈、眼底相機、眼壓計初步篩查眼病和眼壓值,疑似病例應用頻域光學相干斷層掃描(spectral-domain optical coherence tomography,SD-OCT)對視網膜病變進行確診。結果 血糖篩查中發(fā)現4例食蟹猴,3例獼猴血糖升高,眼底未發(fā)現類似人類糖尿病視網膜病變的相關病理改變。13只食蟹猴和6只獼猴出現不同程度的晶狀體混濁。食蟹猴眼壓均值為右眼(15.98±3.315)mm Hg(1 mm Hg=0.133 k Pa),左眼(15.82±4.021)mm Hg;獼猴眼壓右眼(14.9±3.867)mm Hg,左眼(14.7±3.329)mm Hg。1例食蟹猴出現高眼壓,眼底有早期青光眼改變。眼底圖像篩查中,發(fā)現2例食蟹猴眼底出現散在玻璃膜疣分布于視網膜周圍部。1例食蟹猴單眼出現地圖樣脈絡膜病變。結論 在老年猴篩查中發(fā)現了自發(fā)形成的晶狀體混濁、視網膜玻璃膜疣以及脈絡膜萎縮等年齡相關性眼病,證實了靈長類動物與人類有著相似的病理生理改變。同時未見與人類相似的糖尿病視網膜病變眼底改變,可能由于靈長類動物因高血糖引起的微血管結構改變與人類不同,或糖尿病尚未產生視網膜病變,為年齡相關性眼病靈長類動物模型提供了依據。
[Abstract]:Objective to screen natural cataract, age-related macular degeneration, diabetic retinopathy and other eye diseases in the elderly monkeys, and to compare the changes of primate fundus diseases with those of human fundus diseases. To explore the feasibility of establishing an experimental animal model of age-related ophthalmopathy among the elderly monkeys, 39 crab eating monkeys with an average age of 11.9 鹵2.05 years, a minimum of 9 years, a maximum of 17 years and 13 macaques, with an average age of 15.5 鹵5.2 years, a minimum of 7 years, and a maximum age of 24 years, were used. Fasting blood glucose and 2 h glucose tolerance were measured. Slit lamp, fundus camera and IOP were used to screen IOP and IOP. Retinopathy was diagnosed by frequency-domain optical coherence tomography (optical) optical coherence tomographygraphy (SD-OCTT). Results in the blood glucose screening, 4 cases of macaque macaque were found to have elevated blood glucose, and 3 cases of macaque macaques were found to have elevated blood glucose. 13 macaques and 6 macaques had different degrees of lens opacity. The mean IOP of the macaques was 15.98 鹵3.315 mm Hg(1 Hg=0.133 k Pag and 15.82 鹵4.021 mm Hgof the left eye. The intraocular pressure was 14.9 鹵3.867mm Hg in the right eye, 14.7 鹵3.329mm in the left eye and 14.7 鹵3.329mm in the left eye. There are early glaucoma changes in the fundus. It was found that 2 cases of macaque macaque were scattered in vitreous membrane verruca around the retina. 1 case of monocular choroidal lesion was found in 1 case. Conclusion spontaneous lens opacification was found in the screening of senile monkey. Age-related ophthalmopathy, such as vitreous verruca and choroidal atrophy, has proved that primates have similar pathophysiological changes to human beings. It is possible that the changes of microvascular structure caused by hyperglycemia in primates are different from those in humans, or that retinopathy has not been produced in diabetes mellitus, which provides evidence for age-related eye disease primate models.
