大鼠青光眼濾過泡瘢痕化模型中MMP-2及TIMP-2表達的實驗研究

【摘要】:研究目的:建立大鼠青光眼結膜濾過泡瘢痕化模型:觀察結膜下組織瘢痕化過程中基質金屬蛋白酶-2(Matrix metalloproteinase-2, MMP-2)及其組織金屬蛋白酶抑制物一2(tissue inhibitors of Metalloproteinase-2, TIMP-2)的表達情況,初步探討MMP-2及TIMP-2在瘢痕化形成中的作用。 方法:選用健康成年SD大鼠60只,通過前房引流管植入術,成功建立青光眼濾過泡瘢痕化模型。術后觀察濾過泡的形成情況,并分別于術后1d、3d、5d、7d、14d、28d取結膜濾過區(qū)組織分別進行HE染色和western blot檢測,觀察各時間點MMP一2及TIMP-2的表達情況。 結果:1.所有術眼在術后第1天均能形成不同程度隆起明顯的濾過泡。濾過泡維持時間為7-17天,平均生存時間是12天;2.HE染色發(fā)現(xiàn):濾過泡瘢痕形成的過程中可見炎癥細胞、成纖維細胞、血管形成及膠原纖維形成等一系列動態(tài)變化:3.半定量的western blot統(tǒng)計學結果顯示:MMP-2在術后第1天表達降低,術后第3天增高,持續(xù)增高至術后第7天達峰值,隨后逐漸降低至術后第28天,但仍高于對照組,差異具有顯著統(tǒng)計學意義(P0.01)。TIMP-2在術后第1天開始增高,持續(xù)增高至術后第5天達峰值,隨后逐漸降低至術后第28天,但仍高于對照組,差異具有顯著統(tǒng)計學意義(P0.01)。 結論:在大鼠結膜下組織創(chuàng)傷修復過程中,MMP-2及TIMP-2在結膜下組織中表達量發(fā)生變化,說明二者參與結膜濾過泡瘢痕化修復過程。提示研究TIMP-2對MMP-2的特異性抑制作用,可能為開發(fā)新的抗瘢痕化藥物提供依據(jù)。
[Abstract]:Objective: to establish a rat model of glaucoma and conjunctival bleb scarring: to observe the expression of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-2 (TIMP-2) in subconjunctival tissue scarring, and to observe the expression of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-2 (TIMP-2). To explore the role of MMP-2 and TIMP-2 in scarring formation. Methods: 60 healthy adult SD rats were used to establish the model of glaucoma bleb scarring successfully by anterior chamber drainage tube implantation. The formation of filtering bleb was observed after operation. He staining and western blot detection were performed on the conjunctival filtration area at 1 day, 3 days, 5 days, 7 days, 14 days and 28 days, respectively. The expression of MMP-2 and TIMP-2 were observed at each time point. Results 1. On the first day after operation, all operative eyes could form obvious blebs with different degrees of bulge. The mean survival time was 12 days and the mean survival time was 12 days. 2. The inflammatory cells were found in the process of bleb scar formation. A series of dynamic changes, such as fibroblast, angiogenesis and collagen fiber formation, showed that the expression of western blot decreased on the first day after operation, increased on the third day after operation, and continued to increase to the peak value on the 7th day after operation. Then decreased gradually to 28 days after operation, but still higher than that of the control group. The difference was statistically significant (P 0.01). TIMP-2 began to increase on the first day after operation, continued to increase to the peak value on the 5th day after operation, and then gradually decreased to the 28th day after operation. But still higher than the control group, the difference was statistically significant (P 0.01). Conclusion: the changes of expression of MMP-2 and TIMP-2 in subconjunctival tissue during the process of subconjunctival wound repair in rats indicate that they are involved in the scarring repair process of conjunctival filtration bleb. It is suggested that the specific inhibitory effect of TIMP-2 on MMP-2 may be studied. It may provide evidence for the development of new anti-scar drugs.
