【摘要】:鼻咽癌(Nasopharyngealcarcinoma,NPC)是頭頸部惡性程度較高的腫瘤。主要好發(fā)于我國(guó)及東南亞部分地區(qū),而在我國(guó),廣東、廣西是世界鼻咽癌最高發(fā)的地區(qū),其他鄰近區(qū)域(湖南、江西、福建及云貴川等南方諸。┮渤矢甙l(fā)。其發(fā)生目前認(rèn)為與遺傳因素、病毒因素及環(huán)境因素等有關(guān)。 目的:通過(guò)PCR-RFLP技術(shù)研究CYP1A1和GSTM1的基因多態(tài)性在45例云南省鼻咽癌患者和30例健康人群中的表達(dá),從而來(lái)探討這兩種基因的多態(tài)性與云南省鼻咽癌的遺傳易感性之間的關(guān)系,為探尋鼻咽癌發(fā)病機(jī)制及高危人群的篩選提供實(shí)驗(yàn)室基礎(chǔ)和臨床指導(dǎo)。 材料及方法:收集云南省第四人民醫(yī)院病理科自2004年到2012年經(jīng)組織病理學(xué)確診的45例鼻咽癌患者(初診,未經(jīng)過(guò)放化療治療)的石蠟標(biāo)本組織。收集慢性扁桃體炎的石蠟標(biāo)本和正常健康體檢人群血樣標(biāo)本共30例。通過(guò)聚合酶鏈反應(yīng)技術(shù)分析CYP1A1及GSTM1基因多態(tài)性在鼻咽癌患者和正常人群中的表達(dá)。 結(jié)果: 1.單因素CYP1A1MspI的基因多態(tài)性與鼻咽癌易感性無(wú)關(guān),P=0.811,P〉0.05,兩組差異無(wú)統(tǒng)計(jì)學(xué)意義。 2.單因素CYP1A1Exon7的基因多態(tài)性與鼻咽癌易感性無(wú)關(guān),,P〉0.05,兩組差異無(wú)統(tǒng)計(jì)學(xué)意義。 3.單因素GSTM1的基因多態(tài)性檢測(cè)中,GSTM1的功能型(+)和缺陷型(-)對(duì)比在鼻咽癌患者中差異明顯,P〈0.05,說(shuō)明GSTM1的基因多態(tài)性與鼻咽癌的易感性相關(guān)。 4.聯(lián)合分析CYP1A1與GSTM1基因多態(tài)性與鼻咽癌易感性之間的關(guān)系,CYP1A1MspI的基因多態(tài)性聯(lián)合GSTM1基因多態(tài)性分析與鼻咽癌的易感性,結(jié)果和對(duì)照組間無(wú)顯著差異,P〈0.05,兩組差異無(wú)統(tǒng)計(jì)學(xué)意義。 5.通過(guò)單因素分析吸煙與鼻咽癌易感性的關(guān)系,結(jié)果組間差異不顯著,無(wú)統(tǒng)計(jì)學(xué)意義。 結(jié)論: 1.CYP1A1基因多態(tài)性中單獨(dú)的MspI型基因型、Exon7型基因型均與鼻咽癌的易感性無(wú)關(guān)。 2.GSTM1基因多態(tài)性與鼻咽癌的易感性相關(guān),GSTM1缺失型純合子與CYP1A1Exon7的突變型純合子、雜合子同時(shí)存在可以增加鼻咽癌的易感性。 3.通過(guò)單因素分析吸煙及聯(lián)合分析與CYP1A1與GSTM1多態(tài)性的關(guān)系,發(fā)現(xiàn)在本次試驗(yàn)中其鼻咽癌易感性無(wú)關(guān)。
[Abstract]:Nasopharyngeal carcinoma (Nasopharyngealcarcinoma, NPC) is a highly malignant tumor of head and neck. Mainly occurs in some regions of China and Southeast Asia, while in China, Guangdong, Guangxi is the world's highest incidence of nasopharyngeal carcinoma, adjacent area (Hunan, Jiangxi, Fujian and South Guizhou and Sichuan provinces) were also high. The occurrence and genetic factors, viral factors and environmental factors.
Objective: through the expression of gene polymorphism of CYP1A1 and GSTM1 PCR-RFLP technology in Yunnan Province in 45 cases of nasopharyngeal carcinoma and 30 healthy people, the relationship between genetic polymorphism and susceptibility of the two genes with nasopharyngeal carcinoma in Yunnan Province, and to provide clinical laboratory basis for exploring the pathogenesis and risk screening the population of nasopharyngeal carcinoma.
Materials and methods: 45 cases of nasopharyngeal carcinoma patients from the Department of pathology of Fourth People's Hospital of Yunnan province from 2004 to 2012 with histologically confirmed (newly diagnosed, without chemotherapy) of paraffin embedded specimens. The samples were paraffin embedded specimens and normal healthy people were collected. A total of 30 cases of chronic tonsillitis by polymerase chain reaction analysis expression CYP1A1 and GSTM1 gene polymorphism in patients with nasopharyngeal carcinoma and normal people.
