翼狀胬肉和球結(jié)膜中增殖和凋亡相關(guān)因子表達(dá)情況的研究

【摘要】:目的:研究翼狀胬肉和球結(jié)膜組織中增殖和凋亡相關(guān)因子的表達(dá)情況,探討增殖和凋亡的平衡在翼狀胬肉發(fā)病機(jī)制所起到的作用,為臨床上探尋翼狀胬肉的非手術(shù)治療方法提供理論依據(jù)。 方法:臨床收集手術(shù)切除的翼狀胬肉組織標(biāo)本33例,收集正常球結(jié)膜組織23例作為對(duì)照。經(jīng)過福爾馬林固定,石蠟包埋,切片。HE染色,免疫組織化學(xué)方法檢測(cè)翼狀胬肉和球結(jié)膜組織中PCNA、Ki-67、Bax、Bcl-2、mP53、Caspase-3的表達(dá)情況,并使用TUNEL染色法檢測(cè)翼狀胬肉和正常球結(jié)膜組織中處于凋亡的細(xì)胞。并進(jìn)行統(tǒng)計(jì)學(xué)分析比較。 結(jié)果: PCNA在33例翼狀胬肉組織樣本中24例陽性,23例球結(jié)膜組織中7例陽性(P0.01);Ki-67在翼狀胬肉和球結(jié)膜組織中均有表達(dá),但是在翼狀胬肉組織中陽性細(xì)胞數(shù)平均為14.27±4.43,球結(jié)膜組織總陽性細(xì)胞數(shù)平均為4.26±2.42(P0.01);mP53在33例翼狀胬肉組織樣本中15例陽性,在球結(jié)膜組織中全部為陰性(P0.01);Bcl-2在33例翼狀胬肉組織樣本中全為陽性,在23例球結(jié)膜組織中2例陽性(P0.00);Bax在翼狀胬肉和球結(jié)膜組織中均有表達(dá);Caspase-3在33例翼狀胬肉組織標(biāo)本中14例陽性,在23例球結(jié)膜組織中13例陽性,差異無統(tǒng)計(jì)學(xué)意義(P=0.299);TUNEL染色顯示,翼狀胬肉組織中凋亡細(xì)胞局限在上皮基底層細(xì)胞,而球結(jié)膜組織中整個(gè)上皮層均存在凋亡細(xì)胞。 結(jié)論:細(xì)胞增殖和細(xì)胞凋亡均參與翼狀胬肉發(fā)病,翼狀胬肉中細(xì)胞增殖明顯增強(qiáng),細(xì)胞凋亡明顯減弱,細(xì)胞增殖和凋亡之間平衡的打破導(dǎo)致翼狀胬肉的發(fā)病。
[Abstract]:Objective: to study the expression of proliferative and apoptosis-related factors in pterygium and bulbar conjunctiva, and to explore the role of the balance of proliferation and apoptosis in the pathogenesis of pterygium. To provide theoretical basis for exploring non-operative treatment of pterygium. Methods: 33 specimens of pterygium were collected and 23 cases of normal bulbar conjunctiva were collected as control. The specimens were fixed with formalin, embedded in paraffin, stained with HE. The expression of Bcl-2mP53Caspase-3 in pterygium and bulbar conjunctiva was detected by immunohistochemical method, and the apoptotic cells in pterygium and normal bulbar conjunctiva were detected by TUNEL staining. Results: PCNA was expressed in pterygium and bulbar conjunctiva in 24 out of 33 pterygium tissue samples. However, the average number of positive cells in pterygium was 14.27 鹵4.43 and the total positive cell count in bulbar conjunctiva was 4.26 鹵2.42 P0.01mp53 in 33 pterygium tissues. In the bulbar conjunctival tissue, all the specimens of p0.01 and Bcl-2 were positive in 33 cases of pterygium. The expression of Caspase-3 in pterygium and bulbar conjunctiva was positive in 2 out of 23 cases of bulbar conjunctiva and in 33 cases of pterygium and 13 cases in 23 cases of bulbar conjunctiva. The apoptotic cells in pterygium were confined to the basal layer of epithelium, while apoptotic cells were found in the whole epithelium of bulbar conjunctiva. Conclusion: cell proliferation and apoptosis are involved in the pathogenesis of pterygium. The proliferation and apoptosis of pterygium are significantly increased. The imbalance between cell proliferation and apoptosis leads to the pathogenesis of pterygium.
