【摘要】：目的:探索肝臟X受體(liver X receptor,LXR)在濕性年齡相關(guān)性黃斑變性(age-related macular degeneration,AMD)患者外周血單個(gè)核細(xì)胞(peripheral blood mononuclear cells,PBMCs)的表達(dá)及與血脂代謝的相關(guān)性分析。方法:隨機(jī)選取本院眼科門診濕性AMD患者20例,同時(shí)選取健康志愿者20例作為對(duì)照組。收集年齡、性別等病史資料,通過抽血測(cè)定血清總膽固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、高密度脂蛋白膽固醇(high density lipoprotein cholesterol,HDL)、低密度脂蛋白膽固醇(low density lipoprotein cholesterol,LDL)水平。采用熒光定量PCR(q RT-PCR)的方法檢測(cè)LXRα和LXRβ的m RNA的表達(dá)水平。使用LXR激動(dòng)劑TO901317處理,觀察PBMC中LXRα和LXRβ的變化。采用t檢驗(yàn)、非參數(shù)檢驗(yàn)、Pearson相關(guān)性分析及ROC分析進(jìn)行數(shù)據(jù)統(tǒng)計(jì)分析。結(jié)果:濕性AMD患者中LXRα,LXRβ的m RNA的表達(dá)水平都低于對(duì)照組的表達(dá)水平(t=3.881,P=0.001;t=5.839,P=0.000)。使用LXR激動(dòng)劑TO901317處理后,在PBMC中LXRα的m RNA表達(dá)水平升高,LXRβ表達(dá)無差異。Pearson相關(guān)性分析提示LXRα與TG具有負(fù)性相關(guān)性,而LXRα與TC、HDL、LDL及LXRβ與TC、TG、HDL、LDL均無相關(guān)性。ROC分析得LXRα檢測(cè)結(jié)果的曲線下面積(AUC)為0.824(95%CI=0.676~0.973,P=0.001),LXRβ檢測(cè)結(jié)果的AUC為0.945(95%CI=0.873~1.000,P=0.000)。結(jié)論:LXRα和LXRβ的表達(dá)水平下降提示與濕性AMD疾病的發(fā)生發(fā)展密切相關(guān),可能成為診斷和治療濕性AMD的一個(gè)新靶點(diǎn)。
[Abstract]:Objective: to investigate the expression of liver X receptor blood X receptor (LXR) in peripheral blood mononuclear cells (PBMC) from patients with age-related macular degenerationin patients with wet age-related macular degeneration and its correlation with blood lipid metabolism. Methods: ophthalmic departments were randomly selected from our hospital. 20 cases of AMD were diagnosed. At the same time, 20 healthy volunteers were selected as control group. The expression of m RNA of LXR 偽 and LXR 尾 was detected by taking blood samples from serum total cholesterol cholesterol, triglyceride triglyceride, high density lipoprotein cholesterol, low density lipoprotein cholesterol low density lipoprotein cholesterol cholesterol (LDL). The expression of LXR 偽 and LXR 尾 was detected by fluorescence quantitative PCR(q RT-PCR. Level. Treated with LXR agonist TO901317, The changes of LXR 偽 and LXR 尾 in PBMC were observed. Results: the expression level of m RNA of LXR 偽 -LXR 尾 in patients with wet AMD was lower than that of control group (P < 0.05). After treated with LXR agonist TO901317, the expression level of LXR 偽 -LXR 尾 was significantly lower than that of control group. There was no difference in m RNA expression of LXR 偽 in PBMC. Pearson correlation analysis showed that there was a negative correlation between LXR 偽 and TG. However, there was no correlation between LXR 偽 and LXR 尾 and between LXR 尾 and LXR 尾. The area under the curve of LXR 偽 detection results was 0.82495 CI 0.676 ~ 0.973C ~ (0.001) AUC was 0.945 95CI0.8731.000 ~ 0.0000.0000.Conclusion the decrease of the expression levels of LXR 偽 and LXR 尾 is closely related to the occurrence and development of the disease.
