【摘要】：目的研究發(fā)現(xiàn),異常表達(dá)的熱休克蛋白27(heat shock protein 27,HSP27)與多種腫瘤細(xì)胞的侵襲轉(zhuǎn)移相關(guān),但其在鼻咽癌(nasopharyngeal carcinoma,NPC)侵襲轉(zhuǎn)移過程中的作用機制有待研究。因此,本實驗通過檢測NPC組織HSP27、β-連接素(β-catenin)及波形蛋白(Vimentin)的表達(dá),探討NPC組織中HSP27異常表達(dá)對Wnt/β-catenin信號通路及上皮間質(zhì)轉(zhuǎn)化(epithelial to mesenchymal transition,EMT)的影響。方法收集2013-01-01-2015-12-31�？谑腥嗣襻t(yī)院病理科病理確診,為鼻咽未分化非角化性癌患者54例,免疫組化檢測NPC組織中HSP27、β-catenin和Vimentin的表達(dá)。以維持編碼氨基酸序列不變?yōu)榍疤?構(gòu)建經(jīng)密碼子優(yōu)化的HSP27cDNA質(zhì)粒,瞬時轉(zhuǎn)染人鼻咽癌CNE2細(xì)胞,熒光定量PCR(quantitative reverse transcription PCR,qRT-PCR)和蛋白質(zhì)印跡法檢測轉(zhuǎn)染效果,并運用qRT-PCR和蛋白質(zhì)印跡法檢測轉(zhuǎn)染后CNE2細(xì)胞β-catenin、Vimentin的轉(zhuǎn)錄和表達(dá)水平。結(jié)果HSP27高表達(dá)率為35.2%(19/54),HSP27中-低表達(dá)率為64.8%(35/54)。β-catenin在NPC組織中的胞質(zhì)表達(dá)、膜表達(dá)和異質(zhì)性表達(dá)率分別為42.6%(23/54)、27.8%(15/54)和29.6%(16/54);β-catenin異質(zhì)性表達(dá)模式與胞質(zhì)或胞膜模式病例的HSP27表達(dá)強度差異有統(tǒng)計學(xué)意義;Vimentin在NPC組織中的表達(dá)率為63.0%(34/54),與HSP27的表達(dá)強度存在低度相關(guān),r=0.324,P=0.017。CNE2細(xì)胞在轉(zhuǎn)染密碼子優(yōu)化的HSP27cDNA質(zhì)粒后,Vimentin mRNA的下調(diào)差異有統(tǒng)計學(xué)意義,t=6.461,P=0.003;而β-catenin mRNA的下調(diào)差異無統(tǒng)計學(xué)意義,t=2.177,P=0.161;同時β-catenin和Vimentin的蛋白表達(dá)均呈下降趨勢。結(jié)論 HSP27具有抑制NPC細(xì)胞Vimentin基因表達(dá)的功能,該作用可能與抑制β-catenin表達(dá),從而阻抑Wnt/β-catenin信號通路活化有關(guān)。HSP27可能在調(diào)節(jié)NPC發(fā)生EMT和侵襲轉(zhuǎn)移的過程起到重要作用。
[Abstract]:Objective to find that the abnormal expression of heat shock protein 27 (HSP27) is associated with the invasion and metastasis of various tumor cells, but its role in the invasion and metastasis of nasopharyngeal carcinoma (NPC) remains to be studied. By detecting the expression of HSP27, 尾 -catenin and vimentin Vimentin in NPC tissues, we investigated the effect of abnormal expression of HSP27 on Wnt/ 尾 -catenin signaling pathway and epithelial stromal transition to mesenchymal transition.Methods pathological diagnosis was collected from Haikou people's Hospital from January to 31, 2015-31, 2013-01-2015-12-31. The expression of HSP27, 尾 -catenin and Vimentin in NPC tissues was detected by immunohistochemistry in 54 patients with undifferentiated nonkeratinized nasopharyngeal carcinoma. In order to keep the coding amino acid sequence unchanged, a codon optimized HSP27cDNA plasmid was constructed and transiently transfected into CNE2 cells of nasopharyngeal carcinoma (NPC). Fluorescence quantitative PCR(quantitative reverse transcription PCR qRT-PCR and Western blot were used to detect the transfection effect. QRT-PCR and Western blot were used to detect the transcription and expression level of 尾 -catenin vimentin in transfected CNE2 cells. Results the high expression rate of HSP27 was 35.2g / 54 / 54 HSP27. The expression rate of 尾 -catenin in the cytoplasm of NPC was 64.8%. The HSP27 expression intensity of 尾 -catenin heterogenous expression pattern was significantly different from that of cytoplasm or cell membrane pattern in NPC tissues. The expression rate of vimentin in NPC tissues was 63.034 / 54, which was lower than that of HSP27. The down-regulation of Vimentin mRNA in codon optimized HSP27cDNA plasmid was significantly lower than that in 尾 -catenin mRNA, but the down-regulation of 尾 -catenin mRNA was not significant. The protein expression of 尾 -catenin and Vimentin showed a decreasing trend. Inhibition of Vimentin gene expression in NPC cells. This effect may be related to the inhibition of 尾 -catenin expression, thus inhibiting the activation of Wnt/ 尾 -catenin signaling pathway. HSP27 may play an important role in the regulation of EMT and invasion and metastasis in NPC.
