【摘要】：目的:觀察玻璃體腔注射VAS2870對C57小鼠氧誘導(dǎo)視網(wǎng)膜病變的影響。方法:將新生C57BL/6J小鼠隨機(jī)分為3組,分別為正常對照組、VAS2870注射OIR組和無菌PBS緩沖液注射OIR組。將后兩組小鼠在出生后第7d(P7)至P12置于體積分?jǐn)?shù)為75%±2%的恒定高氧氧箱中以構(gòu)建OIR模型,在P12時(shí)給予幼鼠雙眼玻璃體腔注射VAS2870(0.5μL),另一組幼鼠雙眼注射同等劑量的無菌PBS緩沖液。三組小鼠均在P17時(shí)取右眼行視網(wǎng)膜鋪片和Lectin染色,觀察視網(wǎng)膜中央無血管區(qū)及病理性新生血管的情況;取右眼行視網(wǎng)膜組織定量檢測ROS/RNS含量;取左眼應(yīng)用RT-PCR檢測Nox4 m RNA含量,并應(yīng)用Western-blot測定視網(wǎng)膜組織中VEGF的表達(dá)。結(jié)果:VAS2870注射OIR組小鼠視網(wǎng)膜中央無血管區(qū)面積較無菌PBS緩沖液注射OIR組明顯減少(P0.05),病理性新生血管數(shù)目明顯減少(P0.05);VAS2870注射OIR組小鼠視網(wǎng)膜組織Nox4 m RNA的表達(dá)量明顯低于無菌PBS緩沖液注射組;VAS2870注射OIR組小鼠視網(wǎng)膜組織ROS含量較無菌PBS緩沖液注射組明顯降低(P0.05);VAS2870注射OIR組小鼠視網(wǎng)膜組織VEGF的表達(dá)明顯低于無菌PBS緩沖液注射組(P0.05)。結(jié)論:在小鼠OIR模型中,VAS2870可抑制Nox4 m RNA的表達(dá),減少ROS/RNS,下調(diào)VEGF的生成,在視網(wǎng)膜病變進(jìn)程中具有保護(hù)作用。
[Abstract]:Objective: to observe the effect of intravitreal injection of VAS2870 on oxygen-induced retinopathy in C57 mice. Methods: newborn C57BL / 6J mice were randomly divided into 3 groups. The mice in the latter two groups were placed in a constant hyperoxia box with volume fraction of 75% 鹵2% on the 7th day after birth and in a constant hyperoxia box with a volume fraction of 75% 鹵2% to establish the OIR model, and the control group were injected with VAS2870 and aseptic PBS buffer respectively, and the mice in the latter two groups were placed in a constant hyperoxic box with a volume fraction of 75% 鹵2%. VAS2870(0.5 渭 L was injected into the vitreous body of the young rats at P12, and the same dose of aseptic PBS buffer was injected into the eyes of the other group. The right eyes of the three groups were taken out at P17 for retina preparation and Lectin staining. The contents of ROS/RNS in the retinal tissue of the right eye and the content of Nox4 m RNA in the left eye were measured by RT-PCR. Western-blot was used to detect the expression of VEGF in retina. Results the area of central retinal vascular area of mice injected with OIR was significantly lower than that of OIR group injected with sterile PBS buffer solution, and the number of pathological neovascularization was significantly decreased after injection of OIR with VAS2870. The expression of Nox4 m RNA in retina of mice in the control group was significantly lower than that in the control group injected with aseptic PBS buffer solution, and the content of ROS in the retina of the mice injected with OIR was significantly lower than that in the group injected with aseptic PBS buffer solution, and the VEGF in retina tissue of the OIR group was significantly lower than that of the group injected with aseptic PBS buffer. Conclusion: VAS2870 can inhibit the expression of Nox4 m RNA in mouse OIR model. Reducing RNSs and down-regulating the production of VEGF has a protective effect in the process of retinopathy.
