BRMS1基因在不同轉(zhuǎn)移習(xí)性的人鼻咽癌細(xì)胞株中表達(dá)水平的研究

【摘要】:目的:探討不同轉(zhuǎn)移潛能的人鼻咽癌細(xì)胞株中BRMS1基因在mRNA和蛋白表達(dá)水平的差異,并為課題組下一步動(dòng)物實(shí)驗(yàn)提供細(xì)胞理論基礎(chǔ)。 方法:體外培養(yǎng)5株不同組織學(xué)類型及不同轉(zhuǎn)移習(xí)性的人鼻咽癌細(xì)胞株CNE-1、CNE-2Z、SUNE-1、SUNE-1-5-8F、SUNE-1-6-10B。1.應(yīng)用實(shí)時(shí)熒光定量PCR法(real-time PCR)檢測5種人鼻咽癌細(xì)胞株中BRMS1基因mRNA的表達(dá)水平;2.免疫組化和Western blot兩種方法檢測5種人鼻咽癌細(xì)胞株中BRMS1蛋白的定性及定量表達(dá)水平。 結(jié)果:1.應(yīng)用實(shí)時(shí)熒光定量PCR法檢測,發(fā)現(xiàn)不同轉(zhuǎn)移習(xí)性的人鼻咽癌細(xì)胞均擴(kuò)增出BRMS1mRNA,且隨著細(xì)胞轉(zhuǎn)移習(xí)性增高,BRMS1mRNA表達(dá)水平逐漸降低(p0.05);2.通過Western-blot技術(shù)檢測了BRMS1蛋白在人鼻咽癌細(xì)胞中的表達(dá),結(jié)果顯示,BRMS1基因隨著細(xì)胞轉(zhuǎn)移習(xí)性增高,蛋白表達(dá)水平逐漸降低(p0.05);3.通過免疫組織化學(xué)方法檢測了BRMS1在鼻咽癌細(xì)胞中的表達(dá),結(jié)果表明BRMS1在鼻咽癌細(xì)胞膜和/或細(xì)胞質(zhì)中表達(dá)。 結(jié)論:1. BRMS1基因在不同人鼻咽癌細(xì)胞株中mRNA水平和蛋白水平均有表達(dá)且表達(dá)具有一致性,,隨著BRMS1基因mRNA水平表達(dá)的增高,在蛋白水平表達(dá)也呈增高趨勢(shì);2.BRMS1基因在mRNA水平隨鼻咽癌細(xì)胞株轉(zhuǎn)移潛能增高表達(dá)量下降;3.BRMSI基因在蛋白水平隨鼻咽癌細(xì)胞株轉(zhuǎn)移潛能增高表達(dá)量下降;4.BRMS1基因在低轉(zhuǎn)移性細(xì)胞株中高表達(dá),而在高轉(zhuǎn)移性細(xì)胞中低表達(dá),且其表達(dá)強(qiáng)度隨細(xì)胞株的轉(zhuǎn)移潛能的增加而降低,提示了BRMS1可能在鼻咽癌腫瘤細(xì)胞轉(zhuǎn)移機(jī)制中發(fā)揮重要的抑制作用。
[Abstract]:Aim: to investigate the expression of BRMS1 gene in human nasopharyngeal carcinoma (NPC) cell lines with different metastatic potential and to provide a theoretical basis for further animal experiments. Methods: five human nasopharyngeal carcinoma cell lines CNE-1, CNE-2Z, SUNE-1-5-5-8 FT-SUNE-1-6-6-10B.1.The expression of BRMS1 gene mRNA in 5 human nasopharyngeal carcinoma cell lines was detected by real-time PCR method. The expression level of BRMS1 gene was detected by real-time PCR in 5 human nasopharyngeal carcinoma cell lines. The qualitative and quantitative expression levels of BRMS1 protein in 5 human nasopharyngeal carcinoma cell lines were detected by Western blot. Results 1. Real time fluorescence quantitative PCR method was used to detect, It was found that BRMS1 mRNAs were amplified from human nasopharyngeal carcinoma cells with different metastatic habits, and the expression level of BRMS1 mRNA decreased gradually with the increase of metastasis habit. The expression of BRMS1 protein in human nasopharyngeal carcinoma cells was detected by Western-blot technique. The results showed that the protein expression level of BRMS1 gene decreased gradually with the increase of cell metastasis habit. The expression of BRMS1 in nasopharyngeal carcinoma cells was detected by immunohistochemical method. The results showed that BRMS1 was expressed in the cell membrane and / or cytoplasm of nasopharyngeal carcinoma. Conclusion the expression of BRMS1 gene in different human nasopharyngeal carcinoma cell lines is consistent with the expression of mRNA and protein, and the expression of mRNA gene is consistent with the increase of mRNA level of BRMS1 gene. The expression of BRMS1 gene in mRNA decreased with the metastasis potential of nasopharyngeal carcinoma cell line. 3. BRMSI gene expression decreased with the increase of metastasis potential in nasopharyngeal carcinoma cell line at protein level. High expression in migration cell lines, However, the expression of BRMS1 in high metastatic cells decreased with the increase of metastatic potential, suggesting that BRMS1 may play an important role in the mechanism of metastasis of nasopharyngeal carcinoma.
