【摘要】：目的建立斑馬魚(yú)側(cè)線神經(jīng)丘的順鉑損傷模型,檢測(cè)STAT3基因的表達(dá)情況,探討其在毛細(xì)胞順鉑損傷過(guò)程中所發(fā)揮的作用,為抗順鉑耳毒性的研究提供理論依據(jù)。方法將受精后5天的斑馬魚(yú)幼魚(yú)利用250μM順鉑溶液浸泡3小時(shí)建模,通過(guò)Yo-pro-1熒光染色、TUNEL染色了解毛細(xì)胞損傷情況,利用RT-q PCR、原位雜交等技術(shù)檢測(cè)STAT3基因的表達(dá)改變。結(jié)果使用250μM順鉑溶液浸泡斑馬魚(yú)幼魚(yú)3小時(shí),可使其側(cè)線神經(jīng)丘毛細(xì)胞數(shù)降低(P0.001),可誘導(dǎo)側(cè)線神經(jīng)丘出現(xiàn)細(xì)胞凋亡(P0.001),且在此過(guò)程中,STAT3基因的表達(dá)出現(xiàn)上升(P0.001)。結(jié)論STAT3在順鉑對(duì)斑馬魚(yú)側(cè)線神經(jīng)丘毛細(xì)胞的損傷過(guò)程中可能與毛細(xì)胞凋亡相關(guān)并扮演一定角色。STAT3表達(dá)的升高與細(xì)胞凋亡的升高同時(shí)發(fā)生,表明STAT3在一定條件下可能存在促凋亡的作用。
[Abstract]:Objective to establish a model of cisplatin damage in lateral colliculus of zebrafish, to detect the expression of STAT3 gene and to explore its role in the process of cisplatin injury in hair cells. Methods the juvenile zebrafish were soaked in 250 渭 M cisplatin solution for 3 hours after fertilization, and the damage of hair cells was detected by Yo-pro-1 fluorescence staining and Tunel staining. The expression of STAT3 gene was detected by RT-q PCR and in situ hybridization. Results the juvenile zebrafish were immersed in 250 渭 M cisplatin solution for 3 hours. It can reduce the number of hair cells of lateral line colliculus, induce apoptosis of lateral colliculus and increase the expression of STAT3 gene during this process. Conclusion the damage process of STAT3 in Cisplatin on lateral line nerve colliculus hair cells of zebrafish is obvious. [WT5 "HZ] [WT5BZ] [WT5" BZ] [WT5BZ] the results show that the expression of STAT3 gene in the lateral colliculus of zebrafish is damaged by STAT3. The increased expression of STAT3 may be associated with and play a role in hair cell apoptosis. The results suggest that STAT3 may induce apoptosis under certain conditions.
