尼妥珠單抗對(duì)鼻咽癌CNE-2細(xì)胞作用的拉曼光譜研究

【摘要】:目的應(yīng)用拉曼光譜技術(shù)研究尼妥珠單抗?jié)舛群蜁r(shí)間因素對(duì)鼻咽癌CNE-2細(xì)胞作用效果的影響。 方法培養(yǎng)并收集生長(zhǎng)狀態(tài)良好的人鼻咽癌CNE-2細(xì)胞,接種于四個(gè)細(xì)胞培養(yǎng)瓶中。向4個(gè)培養(yǎng)瓶中分別加入0,40ug/ml,150ug/ml,200ug/m四種濃度的尼妥珠單抗注射液各2ml,置于細(xì)胞培養(yǎng)箱中培養(yǎng)24h。采用Renishaw激光顯微共焦拉曼光譜儀對(duì)四種藥物濃度培養(yǎng)的CNE-2細(xì)胞進(jìn)行測(cè)量。每個(gè)藥物濃度組測(cè)量20個(gè)細(xì)胞,將測(cè)得的光譜進(jìn)行PCA-LDA統(tǒng)計(jì)學(xué)分析,尋找最佳藥物作用濃度。再將CNE-2細(xì)胞接種于另外三個(gè)培養(yǎng)瓶中,分別加入最佳藥物濃度的尼妥珠單抗注射液各2ml,置于細(xì)胞培養(yǎng)箱中分別培養(yǎng)24h、48h、72h后進(jìn)行拉曼光譜測(cè)量。 結(jié)果尼妥珠單抗注射液作用24h后,40ug/ml組的CNE-2細(xì)胞平均拉曼光譜與對(duì)照組無(wú)明顯差別,150ug/ml組部分譜峰出現(xiàn)差別,200ug/ml組多個(gè)譜峰出現(xiàn)明顯差別。經(jīng)過(guò)PCA-LDA統(tǒng)計(jì)分析,隨著藥物濃度的提高,藥物組與對(duì)照組的分離趨勢(shì)越發(fā)明顯,200ug/ml時(shí),藥物組和對(duì)照組能夠完全區(qū)分。40ug/ml,150ug/ml,200ug/ml時(shí)判別的靈敏度分別為60%、85%、100%;特異度分別為70%、70%和100%。200ug/ml藥物濃度下,24h組、48h組、72h組的拉曼光譜表現(xiàn)出明顯的差異,但并未表現(xiàn)出與作用時(shí)間相關(guān)的變化趨勢(shì) 結(jié)論拉曼光譜分析表明,在24h作用時(shí)間下,尼妥珠單抗對(duì)CNE-2細(xì)胞的作用效果隨著藥物濃度增加而增強(qiáng),200ug/ml時(shí)的作用效果最為顯著。在相同藥物濃度下,藥物作用效果與時(shí)間梯度未表現(xiàn)出明顯的相關(guān)關(guān)系,可能與譜峰間的相互干擾有關(guān),有待方法學(xué)的改進(jìn)加以明確。
[Abstract]:Objective to investigate the effects of concentration and time of Nitozhu-McAb on nasopharyngeal carcinoma (NPC) CNE-2 cells by Raman spectroscopy. Methods Human nasopharyngeal carcinoma (NPC) CNE-2 cells in good growth state were cultured and inoculated into four culture flasks. Each of the four concentrations of Nietozumab injection was 2ml / 200ugrm. Cultured in cell incubator for 24 hours, CNE-2 cells cultured with four drug concentrations were measured by Renishaw laser microconfocal Raman spectrometer. 20 thin cells were measured in each drug concentration group. The cypress. The spectrum was analyzed by PCA-LDA statistics to find the best drug concentration. Then the CNE-2 cells were inoculated into the other three culture flasks. The optimal concentration of Nietozumab injection was 2 ml each and cultured in a cell incubator for 24 h or 48 h for 72 h. The Raman spectra were measured. Results there was no significant difference in the average Raman spectra of CNE-2 cells between the 40 g / ml group and the control group after 24 hours of treatment with Niytozumab injection. There was no significant difference in some peaks between the 150ug-ml group and the control group. After PCA-LDA statistical analysis, with the increase of drug concentration, the separation trend between the drug group and the control group became more and more obvious. At 200ugrml, the sensitivity of the drug group and the control group was 60% or 85%, respectively. Under the same drug concentration, there was no obvious correlation between the effect and the time gradient, and the effect might be related to the interference between the spectrum peaks. Improvements in methodology remain to be clarified.
