實驗性外斜視貓內(nèi)直肌IGF-1、Anexin A1的表達(dá)變化

【摘要】:目的觀察斜視貓模型弱側(cè)眼外肌中IGF-I及Annexin A1的表達(dá)變化,探討IGF-I、Annexin A1與共同性外斜視發(fā)生、發(fā)展的關(guān)系。 方法3-4周齡幼貓15只,隨機分為實驗組(9只)和對照組(6只)。實驗組貓均佩戴貓類動物用三棱鏡眼鏡。角膜映光法測量斜視度數(shù),確定斜視模型建立。根據(jù)實驗貓佩戴三棱鏡眼鏡持續(xù)時間又分為A、B、C三組,其中A組戴鏡1個月,B組戴鏡2個月,C組戴鏡3個月。分別于佩戴三棱鏡眼鏡1、2、3個月后取實驗組及同齡對照組貓內(nèi)直肌,HE染色觀察內(nèi)直肌形態(tài)學(xué)表現(xiàn),免疫組織化學(xué)染色,測(?)IGF-1、AnnexinA1在眼外肌中的平均光密度值。t檢驗統(tǒng)計比較各實驗組及其同齡對照組組間平均光密度值,單因素方差分析比較實驗組、對照組組間IGF-1、Annexin A1蛋白表達(dá)差異。 結(jié)果①實驗組貓IGF一1的表達(dá)較對照組降低,平均光密度值分別為0.300±0.012、0.265±0.038及0.195±0.028。與同齡對照組比較均有統(tǒng)計學(xué)差異(P0.05)。②實驗組貓AnnexinAl的表達(dá)顯著低于同齡對照組,平均光密度值分別為0.235±0.015、0.185±0.024及0.114±0.017。與對照組比較均有顯著統(tǒng)計學(xué)差異(P0.01)。③實驗組內(nèi)直肌IGF-1、AnnexinA1平均光密度值隨戴鏡時間延長逐漸降低,組間比較有統(tǒng)計學(xué)差異(P0.05)。④各對照組內(nèi)直肌IGF-1、 AnnexinA1表達(dá)無明顯變化,組間比較無統(tǒng)計學(xué)差異(P0.05)。 結(jié)論實驗性外斜視貓弱側(cè)眼外肌中IGF-I、Annexin A1表達(dá)的降低,可能參與共同性外斜視的發(fā)病。實驗性外斜視貓弱側(cè)眼外肌中IGF一1、Annexin A1的表達(dá)隨戴鏡時間延長逐漸降低,可能與共同性外斜視的發(fā)展相關(guān)。佩戴三棱鏡眼鏡能成功誘導(dǎo)共同性外斜視,可作為共同性斜視動物模型的制備方法。
[Abstract]:Objective to observe the expression of IGF-I and Annexin A1 in the weak lateral extraocular muscle of strabismus cat model, and to explore the occurrence of IGF-I Annexin A1 and concomitant exotropia. The relationship between development Methods Fifteen kittens aged 3-4 weeks were randomly divided into two groups: experimental group (n = 9) and control group (n = 6). All cats in the experimental group wore prism glasses and the squint degree was measured by corneal enantiometry. According to the duration of prism glasses, the cats were divided into three groups: group A wearing glasses for 1 month and group B wearing glasses for 2 months. Group C wore glasses for 3 months. After 3 months of wearing prism glasses, the experimental group and the control group were stained with HE staining to observe the morphology of the rectus muscle. The mean optical density of IGF-1 Annexin A1 in the extraocular muscle was compared with the average optical density of the experimental group and the control group of the same age. The single factor variance analysis was used to compare the experimental group. The expression of IGF-1 Annexin A1 protein was different between the control group and control group. Results 1 the expression of IGF-1 in the experimental group was lower than that in the control group, and the average optical density was 0. 300 鹵0. 012, respectively. 0.265 鹵0.038 and 0.195 鹵0.028 respectively. 2 the expression of AnnexinAl in the experimental group was significantly lower than that in the control group. The average optical density was 0.235 鹵0.015. 0.185 鹵0.024 and 0.114 鹵0.017.The difference was significant compared with the control group (P 0.01). 3. IGF-1 of the medial rectus muscle in the experimental group was significantly different from that in the control group. The mean optical density of AnnexinA1 decreased gradually with the prolongation of the time of wearing the lens. There was a significant difference between the two groups (P 0.05). 4. The IGF-1 of rectus muscle in each control group was significantly lower than that in the control group. There was no significant change in AnnexinA1 expression and there was no statistical difference between the two groups (P 0.05). Conclusion the expression of IGF-I and Annexin A1 in the weak lateral extraocular muscles of experimental exotropia cats is decreased. The expression of IGF 1 and Annexin A1 in the weak lateral extraocular muscles of experimental exotropia cats decreased with the prolongation of the time of wear. It may be related to the development of concomitant exotropia. Wearing prism glasses can induce concomitant exotropia successfully and can be used as a preparation method of concomitant exotropia animal model.
