實(shí)驗(yàn)性外斜視貓內(nèi)直肌M-cadherin和CNTF表達(dá)

【摘要】:目的觀察外斜視貓模型中內(nèi)直肌M-鈣粘蛋白(M-cadherin)和睫狀神經(jīng)營(yíng)養(yǎng)因子(CNTF)的表達(dá)變化以及與戴鏡時(shí)間的關(guān)系,探討共同性外斜視的發(fā)病機(jī)制。 方法選擇處于2-4周齡的幼貓15只,雌雄不限,隨機(jī)分為實(shí)驗(yàn)組(9只)和對(duì)照組(6只),實(shí)驗(yàn)組佩戴底向外三棱鏡眼鏡制作外斜視模型,根據(jù)戴鏡時(shí)間分為A、B、C三組,每組3只;對(duì)照組也相應(yīng)分為3組,每組2只。分別于戴鏡后1個(gè)月、2個(gè)月、3個(gè)月取內(nèi)直肌,免疫組化SP法測(cè)量M-cadherin和CNTF的光密度值,統(tǒng)計(jì)分析實(shí)驗(yàn)組與對(duì)照組以及各實(shí)驗(yàn)組間的表達(dá)差異。 結(jié)果實(shí)驗(yàn)組外斜視貓內(nèi)直肌M-cadherin平均光密度值均較對(duì)照組低,A組和對(duì)照A組平均光密度值分別為0.0291±0.0073和0.0386±0.0074,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);B組和對(duì)照B組的平均光密度值為0.0252±0.0046和0.0379±0.0072,C組和對(duì)照C組的平均光密度值為0.0206±0.0035和0.0386±0.0057,兩組差異均有統(tǒng)計(jì)學(xué)意義(P0.05);經(jīng)方差分析,實(shí)驗(yàn)組三組組間差異有統(tǒng)計(jì)學(xué)意義(P0.05),而對(duì)照組組間無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。實(shí)驗(yàn)組CNTF的平均光密度值較對(duì)照組低,A、B、C三組分別為0.364±0.0170、0.328±0.0180和0.277±0.0207,對(duì)照組分別為0.383±0.0172、0.384±0.0117和0.380±0.0273,除A組無(wú)統(tǒng)計(jì)學(xué)意義外(P0.05),B、C兩組差異均有統(tǒng)計(jì)學(xué)意義(P0.05);A、B、C三組組間差異也有統(tǒng)計(jì)學(xué)意義(p0.05),而對(duì)照組組間無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。 結(jié)論外斜視模型內(nèi)直肌中(?)-cadherin和(?)CNTF表達(dá)降低,可能與共同性外斜視的發(fā)生有關(guān)。外斜視模型內(nèi)直肌中CNTF的表達(dá)隨著戴鏡時(shí)間的延長(zhǎng)逐漸降低,可能是共同性外斜視眼外肌失神經(jīng)病理改變逐漸加重的因素。
[Abstract]:Objective to observe the expression of M-cadherin (M-cadherin) and ciliary neurotrophic factor (CNTF) in rectus rectus muscle and the relationship between the expression of M-cadherin and the time of endoscope in exotropia cat model. To explore the pathogenesis of concomitant exotropia. Methods Fifteen kittens aged 2-4 weeks were randomly divided into experimental group (n = 9) and control group (n = 6). According to the time of wearing glasses, they were divided into three groups: group A, group B (n = 3) and group C (n = 3). The control group was also divided into 3 groups, 2 rats in each group, and the optical density of M-cadherin and CNTF were measured by immunohistochemical SP method after one month, two months and three months after wearing the mirror. The difference of expression between the experimental group and the control group was analyzed statistically. Results the average optical density of M-cadherin in the experimental group was lower than that in the control group. The average optical density of group A and control group A were 0.0291 鹵0.0073 and 0.0386 鹵0.0074, respectively. There was no significant difference between group A and group A (P 0.05). The average optical density of group B and control group B were 0.0252 鹵0.0046 and 0.0379 鹵0.0072 respectively. The average optical density of group C and C was 0.0206 鹵0.0035 and 0.0386 鹵0.0057, respectively. The analysis of variance showed that the difference among the three groups in the experimental group was statistically significant, but there was no significant difference between the control group and the control group. The average optical density of CNTF in the experimental group was lower than that in the control group. The values of the three groups were 0.364 鹵0.0170, 0.328 鹵0.0180 and 0.277 鹵0.0207, respectively, and those of the control group were 0.383 鹵0.0172, respectively. 0.384 鹵0.0117 and 0.380 鹵0.0273, with the exception of group A, there was no statistical difference between group A and group C (P 0.05). There was also a significant difference among the three groups (P 0.05), but there was no significant difference between the control group (P 0.05) and the control group (P 0.05). Conclusion the medial rectus muscle of the exotropia model. Cudherin and Con? The decreased expression of CNTF may be related to the occurrence of concomitant exotropia. The expression of CNTF in rectus muscle of exotropia model decreases gradually with the prolongation of the time of wearing glasses. It is possible that the pathological changes of the denervated extraocular muscles in concomitant exotropia are gradually aggravated.
