AMPK-SIRT1信號通路在脊髓損傷后神經(jīng)細(xì)胞自噬和細(xì)胞凋亡調(diào)節(jié)中的作用

目的:脊髓損傷(spinal cord injury,SCI)是一種嚴(yán)重中樞神經(jīng)系統(tǒng)損傷,其發(fā)病率和死亡率較高,已成為一個世界性的醫(yī)學(xué)難題。雖然關(guān)于此疾病的治療研究已經(jīng)在動物模型中取得了重要進(jìn)展,但是目前尚無有效促進(jìn)人類脊髓損傷后功能恢復(fù)的措施。脊髓繼發(fā)性損傷機(jī)制的研究是開發(fā)有效治療策略的前提,因此,SCI研究領(lǐng)域的首要任務(wù)是闡明SCI后復(fù)雜的病理學(xué)調(diào)控機(jī)制。目前大量研究表明5’-腺嘌呤核苷酸激活的蛋白激酶(Adenosine 5‘-monophosphate-activated protein kinase,AMPK)/沉默信息轉(zhuǎn)錄調(diào)節(jié)因子1(silent information regulator of transcription 1,SIRT1)信號通路參與多種疾病,例如,糖尿病,脂肪肝和腫瘤等疾病中細(xì)胞自噬的調(diào)控,但是在中樞神經(jīng)損傷中關(guān)于此信號通路的報(bào)道非常少。最近,研究發(fā)現(xiàn)嚙齒類動物SCI后AMPK和SIRT1的表達(dá)異常,但是AMPK-SIRT1通路如何調(diào)控SCI后神經(jīng)細(xì)胞自噬尚不清楚。本研究首先檢測SCI后體內(nèi)和體外模型中AMPK和SIRT1的時間表達(dá)趨勢,進(jìn)一步采用AMP...
