【摘要】：目的通過慢病毒介導(dǎo)的RNA技術(shù)干擾IL-6的表達(dá),探究脊髓頓挫損傷后Caspase-3的表達(dá)變化。方法Allen′s打擊法制備脊髓頓挫傷(SCC)模型,BBB評分確認(rèn)造模成功;免疫組織化學(xué)和Q-PCR技術(shù)定位和定量測定IL-6和Caspase-3的表達(dá)變化;Gene MANIA網(wǎng)站預(yù)測IL-6與Caspase-3的聯(lián)系;慢病毒介導(dǎo)的RNA干擾技術(shù)構(gòu)建IL-6干擾模型后,Q-PCR技術(shù)定量測定Caspase-3的表達(dá)變化。結(jié)果 SCC后SD大鼠BBB評分顯著低于假手術(shù)組(P0.05);IHC結(jié)果顯示:IL-6和Caspase-3均在在脊髓白質(zhì)膠質(zhì)細(xì)胞表達(dá);Q-PCR結(jié)果顯示:IL-6在損傷后12h表達(dá)顯著增加,3d、28d表達(dá)持續(xù)下降;Caspase-3在損傷后12h、3d、28d表達(dá)持續(xù)下降。生物信息學(xué)分析結(jié)果:IL-6和Caspase-3在大鼠體內(nèi)存在共定位關(guān)系。干擾組Caspase-3表達(dá)比空載組顯著降低(P0.05)。結(jié)論在SCC后,抑制IL-6表達(dá)可降低Caspase-3表達(dá)水平。
[Abstract]:Objective to investigate the changes of Caspase-3 expression after spinal cord collapse injury by lentivirus mediated RNA interference with IL-6 expression. Methods the (SCC) model of spinal cord contusion was established by Allen's percussion method, and the BBB score confirmed the success of the model, and the expression of IL-6 and Caspase-3 was located and quantitatively detected by immunohistochemistry and Q-PCR. The relationship between IL-6 and Caspase-3 was predicted by Gene MANIA website, and the expression of Caspase-3 was quantitatively detected by Q-PCR after the IL-6 interference model was constructed by lentivirus mediated RNA interference technique. Results the BBB score of SD rats after SCC was significantly lower than that of sham operation group (P 0.05); IHC). The results of); IHC showed that IL-6 and Caspase-3 were expressed in white matter glial cells of spinal cord. The results of Q-PCR showed that the expression of IL-6 increased significantly at 12 h after injury, decreased continuously on the 28th day, and decreased continuously on the 28th day after injury, and the expression of Caspase-3 continued to decrease on the 12th day and 28th day after injury. The results of bioinformatics analysis showed that there was a co-localization relationship between IL-6 and Caspase-3 in rats. The expression of Caspase-3 in interference group was significantly lower than that in no-load group (P 0.05). Conclusion inhibition of IL-6 expression can decrease the expression of Caspase-3 after SCC.
