遠(yuǎn)華蟾毒精對(duì)小鼠乳腺癌細(xì)胞侵襲的抑制作用及機(jī)制

【摘要】:目的探討遠(yuǎn)華蟾毒精對(duì)小鼠乳腺癌細(xì)胞侵襲的抑制作用及其機(jī)制。方法將培養(yǎng)好的小鼠乳腺癌細(xì)胞4T1隨機(jī)分為A、B、C、D組。A組常規(guī)培養(yǎng);B、C、D組于缺氧條件下(37℃、5%CO、95%N2)進(jìn)行培養(yǎng),同時(shí)C、D組分別加入0.1、0.5μmol/L遠(yuǎn)華蟾毒精進(jìn)行培養(yǎng)。培養(yǎng)0、6、12、24 h采用劃痕實(shí)驗(yàn)檢測(cè)細(xì)胞遷移能力(遷移距離),用Transwell侵襲實(shí)驗(yàn)檢測(cè)細(xì)胞侵襲能力(穿膜細(xì)胞數(shù)),用Western blotting法檢測(cè)細(xì)胞缺氧誘導(dǎo)因子1α(HIF-1α)、基質(zhì)金屬蛋白酶9(MMP-9)及磷酸化的磷脂酰肌醇3-激酶(PI3K)、蛋白激酶B(AKT)蛋白表達(dá)。結(jié)果不同時(shí)點(diǎn)細(xì)胞遷移距離D組C組B組A組,穿膜細(xì)胞數(shù)D組C組B組A組,HIF-1α、MMP-9、p-PI3K、p-Akt蛋白相對(duì)表達(dá)量D組C組B組A組,以上指標(biāo)兩組間比較差異有統(tǒng)計(jì)學(xué)意義(P均0.05)。結(jié)論遠(yuǎn)華蟾毒精可能通過PI3K/AKT通路降低小鼠乳腺癌細(xì)胞HIF-1α、MMP-9的表達(dá),從而抑制細(xì)胞侵襲。
