【摘要】：背景:脊髓損傷能造成各種運動、感覺功能障礙,肌張力異常及病理反射等,臨床上已通過人臍帶間充質(zhì)干細胞移植對脊髓損傷進行治療。目的:探討大鼠脊髓損傷后不同時期局部移植人臍帶間充質(zhì)干細胞修復(fù)脊髓損傷的效果。方法:選取SPF級雄性成年SD大鼠100只,隨機分為5組:空白對照組、單純脊髓損傷組及3,7,21 d后移植組,各20只。后4組利用Allen’s打擊器制作大鼠T10脊髓損傷模型;3,7,21 d后移植組大鼠分別于脊髓損傷后3,7和21 d在損傷部位周圍移植人臍帶間充質(zhì)干細胞。結(jié)果與結(jié)論:與單純脊髓損傷組相比,3,7,21 d后移植組大鼠造模后49 d運動功能評分增加,血清白細胞介素2水平減少,血清白細胞介素10水平增加,脊髓病理損傷明顯改善,且7 d后移植組效果最為明顯。說明人臍帶間充質(zhì)干細胞移植修復(fù)大鼠脊髓損傷,改善大鼠運動功能,且在大鼠脊髓損傷7 d后移植效果最佳。
[Abstract]:Background: spinal cord injury (sci) can cause various motility, sensory dysfunction, muscular dystonia and pathological reflex, and has been treated clinically by transplantation of human umbilical cord mesenchymal stem cells. Aim: to investigate the effect of local transplantation of human umbilical cord mesenchymal stem cells (HMSCs) on spinal cord injury (sci) after spinal cord injury (sci) in rats. Methods: 100 adult SPF male SD rats were randomly divided into 5 groups: blank control group, simple spinal cord injury group and transplantation group (n = 20). The model of T10 spinal cord injury was made by Allen's in the last 4 groups, and the mesenchymal stem cells of human umbilical cord were transplanted in the transplantation group on the 7th and 21st days after spinal cord injury. Results and conclusion: compared with the simple spinal cord injury group, the motor function score increased, the serum interleukin 2 level decreased and the serum interleukin 10 level increased in the transplantation group 21 days after transplantation. The pathological injury of spinal cord was improved obviously, and the effect of transplantation group was the most obvious 7 days later. The results showed that transplantation of human umbilical cord mesenchymal stem cells to repair spinal cord injury and improve motor function of rats was the best after 7 days of spinal cord injury in rats.
