成軟骨預(yù)分化處理對脂肪干細胞在退變椎間盤中再生修復(fù)作用的影響

【摘要】:目的近年來,干細胞移植逐步成為椎間盤退變性疾病治療的研究熱門。退變椎間盤中惡劣的微環(huán)境對移植干細胞的生長、增殖、分化都有影響。近期有研究表明,"預(yù)分化"處理可以增強移植細胞對微環(huán)境的適應(yīng)能力。本研究旨在探究預(yù)分化處理對干細胞在椎間盤微環(huán)境中生物學(xué)行為的影響。方法將大鼠脂肪干細胞(ADSC,adipose-derived stem cell)分為5組,予Ⅱ型膠原(typeⅡcollagen)包埋處理后,分別進行成軟骨分化處理0天(對照組),3,7,10,14天(實驗組);再將各組細胞移植于模擬退變椎間盤的微環(huán)境(酸性,低糖,高滲透壓)培養(yǎng)7天;用細胞計數(shù)試劑盒(CCK-8)檢測各組細胞的增殖情況,用聚合酶鏈?zhǔn)椒磻?yīng)(RT-PCR)、蛋白印記及免疫熒光法檢測干細胞的細胞外基質(zhì)合成情況。結(jié)果一周的分化過程中,預(yù)分化7天的ADSC增殖能力相對最強。功能基因合成方面,D7組和D10組Ⅱ型膠原mRNA的表達量均較對照組增高;D7組的多聚蛋白聚糖mRNA表達量較對照組高。結(jié)論通過本次研究我們得出結(jié)論,成軟骨細胞預(yù)分化處理7至10天的可能促進脂肪干細胞在椎間盤惡劣環(huán)境中的增殖能力和細胞外基質(zhì)合成能力,有利于干細胞對退變椎間盤的再生。
