【摘要】：目的 :探討大鼠觸液神經(jīng)元(cerebrospinal fluid-contacting neurons,CSF-CNs)p75神經(jīng)營(yíng)養(yǎng)因子受體(p75 neurotrophin receptor,p75NTR)在脊髓損傷后的表達(dá)變化。方法 :成年雌性Sprague-Dawley(SD)大鼠36只,按隨機(jī)數(shù)字表法分為正常對(duì)照組(6只)、假手術(shù)組(6只)和脊髓損傷組(24只),脊髓損傷組采用Allen′s打擊模型(10g×3cm)在大鼠脊髓T10段造成急性脊髓損傷,分別于損傷3d、1周、2周、4周后進(jìn)行取材;對(duì)照組不做任何處理,假手術(shù)組只暴露脊髓,不擊傷脊髓。對(duì)各組大鼠運(yùn)動(dòng)功能行BBB評(píng)分,各時(shí)間點(diǎn)取材行病理切片HE染色觀察。取材前48h側(cè)腦室注射霍亂毒素B亞單位與辣根過(guò)氧化物酶復(fù)合物(CB-HRP)特異性標(biāo)記觸液神經(jīng)元。處死大鼠后,取損傷的脊髓節(jié)段10mm,用免疫熒光雙標(biāo)法檢測(cè)觸液神經(jīng)元p75NTR的表達(dá),ImagePro Plus計(jì)數(shù)目標(biāo)神經(jīng)元CB-HRP/p75雙陽(yáng)性細(xì)胞的數(shù)目。結(jié)果 :假手術(shù)組各時(shí)間點(diǎn)BBB評(píng)分均為21.0±0;脊髓損傷組在術(shù)后3d、1周、2周、4周各時(shí)間點(diǎn)BBB評(píng)分分別為3.20±0.81、10.73±1.02、12.48±1.86、13.29±1.93,兩組各時(shí)間點(diǎn)差異均具有統(tǒng)計(jì)學(xué)意義(P0.05)。HE染色可見(jiàn)正常對(duì)照組和假手術(shù)組脊髓組織結(jié)構(gòu)完整,細(xì)胞形態(tài)正常;脊髓損傷組脊髓組織結(jié)構(gòu)紊亂,神經(jīng)元變性壞死,膠質(zhì)細(xì)胞增生,膠質(zhì)瘢痕和脊髓空洞形成。免疫熒光雙標(biāo)示正常對(duì)照組和假手術(shù)組可見(jiàn)少量CB-HRP/p75雙陽(yáng)性細(xì)胞,計(jì)數(shù)分別為5.16±0.55、4.31±0.61,兩組比較差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);脊髓損傷組傷后3d、1周、2周CB-HRP/p75雙陽(yáng)性細(xì)胞數(shù)分別為13.35±1.53、21.68±2.15、16.26±2.09,與正常對(duì)照組和假手術(shù)組比較差異均有統(tǒng)計(jì)學(xué)意義(P0.05),傷后4周時(shí),CB-HRP/p75雙陽(yáng)性細(xì)胞數(shù)為4.83±0.73,與正常對(duì)照組和假手術(shù)組比較差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 :p75NTR可在大鼠脊髓觸液神經(jīng)元中表達(dá),且在脊髓損傷后表達(dá)增加,觸液神經(jīng)元可能通過(guò)p75NTR參與脊髓損傷的修復(fù)過(guò)程。
[Abstract]:Aim: to investigate the expression of p75 neurotrophin receptor p75 NTR in cerebrospinal fluid-contacting neurons (CSF-CNs) after spinal cord injury. Methods: there were 36 adult female Sprague-Dawley (SD) rats. The rats were randomly divided into normal control group (n = 6), sham operation group (n = 6) and spinal cord injury group (n = 24). Acute spinal cord injury (sci) was induced by Allen's model (10 g 脳 3cm). The samples were taken after 3 days and 2 weeks and 4 weeks after injury, while the control group did not do any treatment, and the sham operation group only exposed the spinal cord and did not hurt the spinal cord. The motor function of rats in each group was assessed by BBB, and pathological sections were stained by HE staining at each time point. Cholera toxin B subunit and horseradish peroxidase complex (CB-HRP) were injected into the lateral ventricle 48 hours before the injection. After the rats were killed, the injured spinal cord segment was taken at 10 mm. The expression of p75NTR in the neurons of contact fluid was detected by immunofluorescence double labeling method. The number of CB-HRP / p75 double positive cells in the target neurons was counted by ImagePro Plus. Results: the BBB score of sham operation group was 21.0 鹵0, and that of spinal cord injury group was 3.20 鹵0.81n 10.73 鹵1.02nil 12.48 鹵1.86n 13.29 鹵1.93 at each time point, and the BBB score of spinal cord injury group was 3.20 鹵0.81n 10.73 鹵1.02n 12.48 鹵1.86n 13.29 鹵1.93, respectively. There were significant differences between the two groups at each time point (P0.05) .HE staining showed that the normal control group and HE staining group had statistical significance (P0.05). The spinal cord of sham-operated group was intact, In the spinal cord injury group, the structure of the spinal cord was disordered, the neurons degeneration and necrosis, the proliferation of glial cells, the formation of glial scar and syringomyelia. Immunofluorescence double labeling showed a small number of CB-HRP / p75 double positive cells in normal control group and sham operation group. The number of CB-HRP / p75 double positive cells in spinal cord injury group was 13.35 鹵1.53 鹵21.68 鹵2.15n 16.26 鹵2.09, which was significantly higher than that in normal control group and sham operation group (P0.05), and CB-HRP / p75 double positive cells in spinal cord injury group were significantly higher than those in normal control group and sham operation group at 4 weeks after injury (P0.05), and the number of CB-HRP / p75 double positive cells in spinal cord injury group was 13.35 鹵1.53n 21.68 鹵2.15n 16.26 鹵2.09 respectively (P0.05). The number of double positive cells was 4.83 鹵0.73, which was not significantly different from that of normal control group and sham operation group (P0.05). Conclusion the expression of p75NTR in rat spinal cord contact neurons is increased after spinal cord injury, and the fluid contact neurons may participate in the repair of spinal cord injury through p75NTR.
