【摘要】：目的探索谷甾醇-3-O-葡萄糖苷對瘢痕疙瘩成纖維細胞(keloid fibroblasts,KF)抑制作用的研究。方法 0(對照組),3.125、6.25、12.5、25、50、100μg/m L的谷甾醇-3-O-葡萄糖苷作用KF細胞48 h后,MTT法檢測細胞活力,Ed U染色法檢測細胞增殖,流式細胞術分別檢測細胞凋亡及細胞周期,酶聯(lián)免疫吸附試驗(ELISA)檢測細胞中COL I、COL III含量。結果 3.125、6.25、12.5、25、50、100μg/m L谷甾醇-3-O-葡萄糖苷能顯著的降低KF細胞活力(P0.01),提高細胞增殖抑制率(P0.01),IC50=(27.54±2.18)μg/m L。與對照組比較,12.5、25、50μg/m L谷甾醇-3-O-葡萄糖苷能顯著的降低細胞增殖率(P0.01),提高細胞早期及晚期凋亡率(P0.01),使細胞周期阻滯在G1期(P0.01),同時還能顯著的降低細胞中COL I、COL III含量(P0.01)。結論谷甾醇-3-O-葡萄糖苷能顯著的抑制KF細胞增殖,誘導細胞凋亡,同時還能抑制膠原的合成。
[Abstract]:Objective to investigate the inhibitory effect of sitosterol-3-O- glucoside on keloid fibroblast KF in keloid fibroblasts. Methods the KF cells were treated with sitosterol -3-O- glucoside for 48 h. The proliferation of KF cells was detected by MTT assay. The apoptosis and cell cycle of KF cells were detected by flow cytometry. Enzyme linked immunosorbent assay (Elisa) was used to detect the content of Col III in cells. Results 3.125 ~ 6.25 ~ (12.5) 渭 g / mL sitosterol -3-O- glucoside could significantly reduce the activity of KF cells and increase the cell proliferation inhibition rate (P _ (0.01) ~ (50) 渭 g / m ~ (-1) ~ (27. 54 鹵2. 18) 渭 g / m ~ (-1). Compared with the control group, 12.5g / mL sitosterol -3-Oglucoside could significantly decrease the cell proliferation rate and increase the cell apoptosis rate in the early and late stages of the cell, and make the cell cycle arrest in G1 phase (P0.01A). At the same time, it could significantly reduce the content of Col ICOL III in the cells. Conclusion sitosterol-3-O- glucoside can significantly inhibit the proliferation of KF cells, induce apoptosis and inhibit the synthesis of collagen.
