沉默Aurora-B抑制骨肉瘤細胞143B在裸鼠體內(nèi)生長

【摘要】:目的:初步探討沉默Aurora-B基因?qū)θ斯侨饬黾毎?43B在裸鼠體內(nèi)生長的影響。方法:構(gòu)建人骨肉瘤143B細胞裸鼠移植瘤模型18只,隨機分為3組(n=6):分別為LV-sh Aurora-B(AURKB)注射組(實驗組)、LV Negative注射組(陰性對照組)和PBS注射組(空白對照組)。每3天(間隔2天)于腫瘤組織局部注射(共注射3次)。每7天皮外測量瘤體大小,5周后處死,取出瘤體,測量瘤體大小及質(zhì)量,TUNEL法檢測細胞凋亡指數(shù)。結(jié)果:LV-sh Aurora-B注射組瘤體生長速度顯著低于LV Negative注射組及PBS注射組(P0.05),LV Negative注射組與PBS注射組之間比較無顯著統(tǒng)計學差異;5周后,LV-sh Aurora-B注射組瘤體大小、質(zhì)量顯著低于LV Negative注射組及PBS注射組,LV Negative注射組與PBS注射組之間比較無顯著統(tǒng)計學差異。LV-sh Aurora-B注射組瘤體組織細胞凋亡指數(shù)顯著高于LV Negative注射組及PBS注射組(P0.05),LV Negative注射組與PBS注射組之間比較無顯著統(tǒng)計學差異。結(jié)論:在裸鼠體內(nèi),沉默Aurora-B基因能有效誘導人骨肉瘤143B細胞凋亡,抑制人骨肉瘤143B細胞增殖。
[Abstract]:Aim: to investigate the effect of silencing Aurora-B gene on the growth of human osteosarcoma cell line 143B in nude mice. Methods: eighteen nude mice transplanted with human osteosarcoma 143B cells were randomly divided into three groups: LV-sh Aurora-BU AURKB injection group (experimental group) (negative control group) and PBS injection group (PBS injection group). The tumor tissue was injected locally every 3 days (2 days interval) (3 times). The tumor size was measured every 7 days and killed after 5 weeks. The tumor size and mass were measured and the apoptosis index was detected by Tunel. Results there was no significant difference in tumor growth rate between LV-sh Aurora-B injection group and PBS injection group and between PBS injection group and PBS injection group. After 5 weeks, the tumor size in LV-sh Aurora-B injection group was significantly lower than that in LV-Aurora-B injection group. There was no significant difference between the LV-sh Aurora-B injection group and the PBS injection group. The apoptotic index of the tumor tissue in the LV-sh Aurora-B injection group was significantly higher than that in the LV Negative injection group and the PBS injection group, and the LV-sh Aurora-B injection group was significantly higher than that in the LV Negative injection group and the PBS injection group. There was no significant difference between the ejection group and the PBS injection group. Conclusion: in nude mice, silencing Aurora-B gene can effectively induce apoptosis of human osteosarcoma 143B cells and inhibit the proliferation of human osteosarcoma 143B cells.
