【摘要】：目的 探究環(huán)氧合酶(Cox)-2選擇性抑制劑塞來昔布對大鼠創(chuàng)傷性異位骨化的防治機制。方法 創(chuàng)傷性異位骨化大鼠予塞來昔布治療,分別采取喂食2、6、10 w的不同方案,治療后觀察各組肢體的皮溫、腫脹程度及X線表現(xiàn),并通過實時定量PCR及Western印跡實驗,檢測跟腱組織骨形態(tài)發(fā)生蛋白(BMP)-4的表達含量。結(jié)果 塞來昔布組皮溫、腫脹程度、新骨形成均顯著低于對照(生理鹽水處理)組(P0.05)。同時,隨著塞來昔布處理時間的延長,皮溫均明顯降低、腫脹程度及新骨形成情況均明顯減輕;實時定量PCR及Western印跡檢測顯示:與對照組比較,塞來昔布組跟腱組織BMP-4的表達明顯下調(diào),并且隨時間的延長,BMP-4的表達也相應(yīng)下調(diào)。結(jié)論 塞來昔布可通過降低大鼠跟腱組織BMP-4的表達以干預(yù)異位骨化。
[Abstract]:Objective to investigate the protective mechanism of celecoxib, a selective inhibitor of cyclooxygenase-2, on traumatic ectopic ossification in rats. Methods Rats with traumatic ectopic ossification were treated with celecoxib for 10 weeks. The skin temperature, swelling degree and X ray appearance of each group were observed, and real-time quantitative PCR and Western blot were used. The expression of bone morphogenetic protein (BMP- 4) in Achilles tendon was detected. Results the skin temperature, swelling and new bone formation in celecoxib group were significantly lower than those in control group (P 0.05). At the same time, with the prolongation of celecoxib treatment time, the skin temperature was significantly decreased, the swelling degree and new bone formation were significantly reduced, and real-time quantitative PCR and Western blot analysis showed that: compared with the control group, In celecoxib group, the expression of BMP-4 in Achilles tendon was down-regulated, and the expression of BMP-4 was down-regulated with time. Conclusion Celecoxib can interfere with ectopic ossification by decreasing the expression of BMP-4 in Achilles tendon.
