丙泊酚對腎缺血-再灌注損傷大鼠腎葉間動脈縫隙連接Cx43表達的影響

【摘要】:目的觀察丙泊酚干預對腎缺血再灌注損傷大鼠腎葉間動脈(RIA)Cx43表達的影響。方法雄性SD大鼠隨機分為空白對照(control)4h、24h組,假手術(shù)(sham)4h、24h組,腎缺血再灌注(I/R)4h、24h組,丙泊酚干預(propofol)4h、24h組,脂肪乳劑(intralipid)4h、24h組。采用切除右腎、無創(chuàng)動脈夾夾閉左腎制備腎缺血再灌注模型,腎缺血45min,按分組不同分別再灌注4h、24h;采用全自動生化儀檢測血清尿素氮(serum urea nitrogen,BUN)和肌酐(creatinine,Cr);HE染色法觀察腎組織的病理學變化;壓力肌動圖技術(shù)觀察RIA收縮舒張的變化;Western blot方法檢測RIA Cx43蛋白的表達。結(jié)果與空白對照組相比,假手術(shù)組血清BUN、Cr濃度無統(tǒng)計學差異,腎HE染色未發(fā)現(xiàn)明顯的形態(tài)學改變,壓力肌動圖觀察RIA收縮率無統(tǒng)計學差異,Cx43蛋白表達無統(tǒng)計學差異;與假手術(shù)組相比,腎缺血再灌注組血清BUN、Cr濃度均顯著增高,腎HE切片顯示腎組織病變明顯,壓力肌動圖顯示RIA收縮率下降,Cx43蛋白表達顯著增高,差異有統(tǒng)計學差異(P0.05);與腎缺血再灌注組相比,丙泊酚干預組血清BUN、Cr濃度有所降低,腎HE切片顯示腎組織病變減輕,RIA收縮率有所升高,Cx43蛋白表達降低,差異有統(tǒng)計學意義(P0.05)。結(jié)論丙泊酚可能通過降低RIA Cx43蛋白的表達改變血管舒縮性,從而減輕腎缺血再灌注損傷。
[Abstract]:Objective to observe the effect of propofol on the expression of RIA-Cx43 in renal interlobar artery during renal ischemia-reperfusion injury in rats. Methods male Sprague-Dawley rats were randomly divided into control group (control group), sham group (n = 4), renal ischemia reperfusion group (n = 4), propofol intervention group (n = 24) and fat emulsion intra (n = 4 h) group (n = 24). The model of renal ischemia-reperfusion was established by resection of right kidney and occlusion of left kidney by non-invasive artery clamp. After 45 minutes of renal ischemia, the rats were divided into different groups for 4 hours and 24 hours after reperfusion respectively. The pathological changes of renal tissues were observed by using automatic biochemical instrument to detect serum urea nitrogenbun and creatinine (creatinine). The changes of systolic and diastolic activity of RIA were observed by pressure-actin technique. The expression of RIA Cx43 protein was detected by Western blot. Results compared with the blank control group, there was no significant difference in serum BUNN Cr concentration, no obvious morphological changes in renal HE staining, and no significant difference in the expression of Cx43 protein in the RIA contraction rate observed by pressure-actigraphy. Compared with the sham-operated group, the serum BUNN Cr concentration in the renal ischemia reperfusion group was significantly higher, the renal HE section showed obvious renal tissue lesion, and the pressure actin graph showed that the RIA contraction rate decreased and the expression of Cx43 protein increased significantly. Compared with the renal ischemia-reperfusion group, the serum BUNX Cr concentration in the propofol treated group was decreased, and the renal HE section showed that the renal tissue lesion was alleviated and the RIA contraction rate was increased, the expression of Cx43 protein was decreased, and the difference was statistically significant (P 0.05). Conclusion propofol may reduce renal ischemia-reperfusion injury by decreasing the expression of RIA Cx43 protein.
