【摘要】:目的建立皮膚擴(kuò)張模型,探究皮膚擴(kuò)張?jiān)缙谥衜iRNA和mRNA表達(dá)譜的變化情況。方法取6-8周SD大鼠建立擴(kuò)張模型,在擴(kuò)張?jiān)缙谌∠聰U(kuò)張皮膚,并利用未擴(kuò)張皮膚作為對照,采用Affymetrix microRNA 3.0平臺和Affymetrix GeneChip Rat Gene 1.0 ST檢測擴(kuò)張?jiān)缙趍iRNA和mRNA的表達(dá)變化;利用統(tǒng)計(jì)學(xué)方法初步篩選出表達(dá)發(fā)生變化的miRNA和mRNA,并根據(jù)差異表達(dá)的倍數(shù)篩選出表達(dá)差異較大的關(guān)鍵miRNA。采用TargetScan、miRDB和IPA數(shù)據(jù)庫對關(guān)鍵miRNA進(jìn)行靶點(diǎn)預(yù)測;利用miRNA與靶點(diǎn)mRNA數(shù)量變化呈負(fù)相關(guān)的關(guān)系初步明確擴(kuò)張?jiān)缙趍iRNA可能調(diào)控的mRNA靶點(diǎn),建立miRNA-靶mRNA的一一對應(yīng)關(guān)系。對靶點(diǎn)mRNA進(jìn)行功能分析和通路分析,明確擴(kuò)張?jiān)缙陉P(guān)鍵miRNA-靶mRNA-通路的對應(yīng)關(guān)系。結(jié)果篩選出了11個滿足統(tǒng)計(jì)學(xué)差異和表達(dá)倍數(shù)變化的miRNA,明確了miRNA-靶mRNA之間的相互作用關(guān)系;這些miRNA調(diào)控的靶點(diǎn)參與TGF-βSignaling、Apoptosis Signaling和PDGF Signaling等通路。結(jié)論本研究篩選出了11個皮膚擴(kuò)張?jiān)缙诘年P(guān)鍵miRNA,調(diào)控?cái)U(kuò)張過程中的關(guān)鍵通路,這些miRNA可作為調(diào)控皮膚早期擴(kuò)張的靶點(diǎn)。
[Abstract]:Objective to establish a skin dilatation model and investigate the changes of miRNA and mRNA expression profiles in the early stage of skin dilatation. Methods Sprague-Dawley rats (6-8 weeks old) were used to establish dilatation model. The expression of miRNA and mRNA were detected by Affymetrix microRNA 3.0 platform and Affymetrix GeneChip Rat Gene 1.0ST in the early stage of dilatation. The expression of miRNA and mRNAs were preliminarily screened by statistical method, and the key miRNAs were screened according to the multiple of differentially expressed miRNAs. The target of miRNA was predicted by TargetScanmiRDB and IPA database, and the relationship between miRNA and the quantity of mRNA was negatively correlated, and the mRNA target which might be regulated by miRNA in the early period of expansion was identified, and the one-to-one correspondence of miRNA-target mRNA was established. The functional analysis and pathway analysis of target mRNA were carried out to clarify the relationship between the key miRNA-target mRNA-pathway in the early stage of expansion. Results Eleven miRNAs satisfying the statistical difference and the change of expression multiple were screened, and the interaction between miRNA-target mRNA was clarified, and these miRNA regulatory targets were involved in the pathway of TGF- 尾 signaling Signaling and PDGF Signaling. Conclusion in this study, 11 key miRNAs in the early stage of skin dilatation were screened and the key pathways in the process of skin expansion were regulated. These miRNA can be used as targets for early skin dilatation.
